Objective: This study aimed to systematically studies on chemical compositions of fresh Gastrodia elata Blume,selected their neuroprotective activities in vitro,and founded effective monomer components with neuroprotective activities,which provide theoretical basis for pharmacological research of neuroprotective activity of Gastrodia elata Blume.Methods: Fresh Gastrodia elata Blume extract were filtered by D101 macroporous resin,then separated and purified by gel column chromatography,RP-C18 medium pressure liquid chromatography and semi preparative liquid chromatography.Meanwhile,high speed countercurrent chromatography technology was applied to separate and purify ethanol extract from fresh Gastrodia elata Blume.Structures of isolated monomers were identified by spectral data of 1H-NMR,13C-NMR,MS and other methods;using CCK method to study neuroprotective activity of the monomers.Results: A total of twenty-one compounds were isolated and identified,respectively: p-hydroxybenzylalcohol(1),syringic acid(2),4-hydroxybenzyl methyl ether(3),p-hydroxybenz aldehyde(4),N6-(4-hydroxyzenzyl)adenosine(5),s-(4-hydroxybenzyl)-glutathione(6),Gastrolatathioneine(7),4-hydroxy-3-(4-hydroxybenzyl)benzyl methyl ether(8),4,4’-dihydroxydiphenyl methane(9),p-hydroxybenzyl ethyl ether(10),4,4’-dihydroxydibenzyl ether(11),2,4-bis(4-hydroxybenzal)-phenol(12),Gastrol A(13),1-O-(4-hydroxymethylphenoxy)-3-O-trans-cinnamoyl-β-D-glucoside(14),Uracil(15),adenosine(16),3,4-dihydroxybenzaldehyde(17),3,4-dihydroxy benzyl ethyl ether(18),4-(hydroxymethyl)-1,2-benzenediol(19),4-ethoxymethylphenyl-4’-hydroxybenzyl ether(20),compound 21.Conclusions: Through the study of fresh Gastrodia elata Blume ethanol extracts,we identified twenty-one compounds,including one new compound,two compounds for the first time isolated from Gastrodia elata Blume;We find that six compounds in fresh gastrodia elata have obvious neuroprotective activity and are potential neuroprotective substances through the study of the protective activity of the compound monomers.
