Objective:Topics selected to the TCM righting solid form on behalf of parties Yupingfengpowder to do intervention in asthmatic mice experiments, by observing Yupingfengpowder asthmatic mice in BALF of (bronchoalveolar lavage fluid) of Th17-relatedcytokines (such as: IL-17and TGF-β1and the expression of IL-6) to explore thepossible mechanism of Yupingfeng powder prevention and Treatment of Bronchialasthma, provide new ideas for the clinical application of Yu Ping Feng powder.Methods:1.An established mouse model of asthma:48SPF grade female BALB/C mice wererandomly divided into normal control group, asthmatic model group, Yupingfengpowder high in the low dose group, dexamethasone group: In addition to normalcontrol group, sensitized and challenged with saline instead of the other groups in theexperimental section1d,8d,15d, were injected intraperitoneally the0.2mL sensitizedliquid,22-28d aerosol inhalation of2%of OVA saline; Yupingfeng powder thetreatment group and dexamethasone group were administered30min before theatomization excitation and intraperitoneal injection.2BALF inflammatory cell classification counts; detected by double antibodysandwich ELISA in BALF IL-17, TGF-β1and IL-6levels; lung histopathology HEstaining to observe the gross morphology of the lung tissue and inflammatory cellinfiltration.Results:1.Counting of leukocytes and eosinophils, neutrophils and lymphocytes in the asthmagroup mice in BALF was significantly higher than the normal control group (P <0.05); dexamethasone group, Yupingfeng powder dose neutrophil, eosinophils andlymphocytes was significantly reduced, compared with the asthmatic group differencewas significant (P <0.05).2.Lung tissue pathology observed: the asthmatic mice lung bronchioles andperivascular inflammatory cell infiltration, pulmonary interstitial and alveolar spacesvisible structural disorder in inflammatory cell infiltration, airway, airway epithelialhyperplasia, hypertrophy,bronchial mucosa folds to reduce, or even fall off, stenosisincreased significantly in the lumen of mucus exudate, alveolar septal expansion,thickening of the surrounding tissue edema and inflammatory cell infiltration; clearthe bronchial and alveolar structure of lung tissue of normal miceairway mucosalepithelial integrity, and a small amount of inflammatory cell infiltration.3. Yu Ping Feng Powder asthmatic mice in BALF cytokines IL-17, IL-6, TGF-betaexpression: BALF of asthmatic mice, IL-17, IL-6was significantly increasedcompared with normal group, the difference was significant (P <0.05); dexamethasonegroup, middle dose Yupingfeng powder the group mice BALF IL-17levels decreasedsignificantly with the asthma group difference was significant (P <0.05), promptedYupingfeng powder dexamethasone inhibit asthmatic mice in BALF IL-17;dexamethasone group mice the in BALF in IL-6contents with the asthmatic groupcompare the difference was not of significant significance (P>0.05); YupingfengPowder in high, the, the low-dose group of mice in BALF in of IL-6content of withasthma group compare is still high, but the difference no statistically significant (P>0.05), suggesting that Yupingfeng powder asthmatic mice in BALF IL-6had nosignificant regulatory role.Conclusion:1.IL-17, IL-6involved in the pathogenesis of bronchial asthma.2Yupingfeng powder can reduce asthmatic mice lung bronchioles and perivascularinflammatory cell infiltration, improve the status of airway inflammation, reduceairway epithelial mucus secretion.3Yupingfeng powder asthma mouse acute phase of IL-17inhibition, Yupingfengpowder inhibit Th17cells involved in asthma, this inhibition by upregulating the expression of Th1cells and promote IFN-γ secretion, thereby inhibiting IL-17production.
