As a third generation host compound of supramolecular, calixarene has become ahotspot of research fields. The calixarene has flexible conformation and size-controlled cavitystructure, combined with the molecule itself has multiple reaction sites. Variety of calixareneand its derivatives have been synthesized. Now these derivatives of calixarene havedemonstrated the broad application prospects.such as in the identification of the ion, magneticresonance imaging contrast agents, chemical sensors, anti-bacteria and other biochemically.In this paper, We synthesized two kinds of water-soluble amino calixarene derivatives, and onthis basis, the interaction of the amino calixarene and human serum albumin were discussed.1. In accordance with literature, we synthesized p-t-butylcalix[4]arene and calix[4]arene.2. Start from calix[4]arene, we use diazotization to get5,11,17,23-4[(rightcarboxyphenyl) azo]-calix [4] arene by diazotization, and then we synthesized5,11,17,23-4-amino-calix[4]arene by the tin-hydrochloric acid reduction.3.We get5,11,17,23-4[(diethylamino)-methyl]-calix [4] arene by Mannich reactionof calix[4]arene。And then we use concentrated hydrochloric acid to synthesize5,11,17,23-[(dimethylamino)-methyl]-calix [4] arene hydrochloride.4. Explore the impact on fluorescent quenching in the case of different temperature,concentration, and different amino calixarene derivatives. Then calculated the bindingconstants and number of binding sites in both the formation of complexes. We can determinethe binding of the amino calixarene derivatives to human albumin is the hydrophobiceffect.The result showed that the amino calixarene derivatives can quench the fluorescenceintensity of human albumin,and we found to be in static quenching procedure.
