Brucella is a small invasive, non-motile, gram-negative coccobacillus that multiplies and survives within a broad range of eukaryotic cells. Brucellosis, also known as undulant fever or Mediterranean fever, is caused by this genus of bacteria and is still one of the major, highly transmissible, zoonotic diseases affecting people and animals4kinds of serological tests, as Rose Bengal plane Test (RBPT)、indirect enzyme linked immunosorbent assay (i-ELISA), standard agglutination test (SAT) and competitive enzyme linked immunosorbent assay (c-ELISA) were used to detect6577serum samples from dairy cows on farm where aborted that in the north of China. Positive rate of serological test was12.56%.12relative samples of lung, liver, spleen etc. in aborted fetus of dairy cow were selected to perform bacteriology detection and also by multiplex PCR,AMOS-PCR, MLVA.4pathogenic strains were isolated and cultured.lt was confirmed as brucellosis and identified as B. abortus biovar3.The causes and characteristics of brucellosis outbreak were explored with the background information.The number of positive samples detected by RBPT、i-ELISA、SAT、c-ELISA were543、501、460、500, and the positive rate were62.20%、51.39%、52.69%.57.27%. The RBPT, SAT, i-ELISA and c-ELISA indetecting the infected cattle, almost obtained high consistency rate, but RBPT had higher false positive rate and SAT had higher false negative rate. The data suggested that RBPT screening test could be used in the field, and the positive results should be further confirmed by i-ELISA or c-ELISA.The i-ELISA kit evaluated in this study was thought to be a valuable tool for the diagnosis of bovine brucellosis, especially where large numbers of serum should be tested to obtain such information. The sensitivity and specificity of i-ELISA kits from Ⅰ,Ⅱ,Ⅲ,three companies were evaluated compared with c-ELISA. The results shows that the i-ELISA kit from the Ⅱ company is the most sensitive of all.
