With the rapid development of science and technology, more and more attention hadbeen paid to the electromagnetic radiation. Extremely low frequency electromagnetic fieldis one of the electromagnetic fields that we often encounter in our daily life. As the biggestperipheral immune organ, spleen plays an important role in immune reaction of humanbody. It is also the place where lymphocyte grow, split apart and be mature finally. So thistask observes the spleen lymphocytes of spleen of mice with extremely low frequencyelectromagnetic field, to study effect of electromagnetic to cellular immunity.During the course of the experiment, we bought45Kunming mice and the mice wererandomly divided into3groups,15for each group. We use an extremely low frequencyalternating electromagnetic (0.05mT,50Hz) and an extremely low frequency pulsedelectromagnetic field (0.01mT,50Hz,15s) affect on mice, for five hours a day. After theexperiment, measure the weight of mice, record the data. Mice were killed after six weeks,under aseptic condition, get the mice spleen. Then weigh it up and count the spleenexponent. Calculate the spleen indices of the mice. Grate spleen, after washing3times, andthen cultured in RPMI1640medium, get the spleen cell suspension. Cell suspension isgrown in96-well plates and treated with the appropriate conditions, tissue in CO culturebox. Four hours before the end of the cell culture, take out the96-well plates. Adding theMTT into the Cells suspension, continue to incubate4hours. After the culture end, takeout the96-well plates, adding the DMSO. After a fully coaster, using the spectrophotometer determines the content of OD. In addition, sub the Cells suspension intofive96-well plates. Take out a plate everyday, measure the OD and count the lymphocytes.Cell growth curves are detected by MTT Method and Cell Counting Method.Result showed that extremely low frequency alternating electromagnetic field canpromote weight increased of mouse. Alternating electromagnetic field group indexes ofspleen and lymphocyte proliferation rate are higher than normal control group. Pulsedelectromagnetic field can promote increased indexes of spleen of mice.We can see fromthe lymphocyte growth curve with the extension of incubation time the effect wears off.
