Objective:1.explore the effect of low frequency electromagnetic fields on the proliferration of glioma cells.2.To order to establish C6 rat intracerebral glioma model,investigate rat brain glioma impact by the way of extremely low frequency electromagnetic field exposure.Methods:1.C6 brain malignant glioma cells were cultured in vitro,it were divided to a treatment group and a control group.The C6 cells in the treatment group were exposed in Extremely low frequency electromagnetic field which have 40 Hz and magnetic field strength for 2.6mT.the control group were not exposed to the magnetic field.In the magnetic field after 24h,48h,72h,respectively,collecting specimens,determined by MTT tumor survival,growth curve described and were observed in tumor cell growth state,and it was contrasted with the control group.2.C6 cells were inplanted into all the SD rats,the number is 1×10~6 each rat.SD rat with intracranial C6 gliomas were established using stereotactic methods,then rats were randomly divided into treatment group(n=15) and control group(n=15).From 3 days after tumor inoculation,the rats in treatments group were exposesd three hours in the magnetic field every day,it lasted a week.,the control group were not exposed to magnetic field.during the time,we observe the spirit of rats,neural function (hemiplegia),the survival any time and death in rats with pathological biopsy autopsy.The results:1MTT Experimental results showed that:under the exposure of 40 Hz extremely low frequency electromagnetic field at 2.6mT,the C6 glioma cell growth is slightly inhibited, when a longer exposure time And no significant impact had happened on C6 glioma cell when a relatively short-time exposure to extremely low frequency electromagnetic fields. Significant difference had only existed between the control group and the experimental group under 3h/d exposure,the growth of C6 glioma cells were good through the microscope observing.2 The rats of experimental group died in 17-28 days from the inoculation of C6 glioma cell,with an average survival of 22 days,while the rats of control group had a long-time survival.Significant difference had happened between the experimental group and the control group.(P<0.05).The tumor was founded in the dead rats of experimental group.The pathology confirmed that the tumor founded in the dead rats of experimental group was C6 glioma and the tumor inoculation rate was 100%.3,The rats of control group died in 16-27 days from the inoculation of C6 glioma cell,with an average survival of 22 days,while the rats of experimental group died in 21-33 days from the inoculation of C6 glioma cell,with an average survival of 26 days.Significant difference had happened between the experimental group and the control group.(P<0.05).The C6 glioma were founded both in experimental group and control group.The morphological difference of the C6 glioma between the experimental group and the control group was not founded.Conclusion:1.The short-term exposure of 40 Hz extremely low frequency electromagnetic field at 2.6mT had no significant effect on the growth of C6 glioma cells,while under long-term exposure,the growth of the C6 glioma cells were inhibited.2.The intracranial model of rats was reliable through the 10ul cell injection at the concentration of 1×10~8/ml.3. Extremely low frequency electromagnetic field exposure may be extended survival time of tumor-bearing rats,but it can not cure glioblastoma.Short-term exposure of low-intensity extremely low frequency electromagnetic field on patients with glioma has non- toxic or treatment,but long-term exposure of low-intensity extremely low frequency electromagnetic field may have a therapeutic effect on patients with glioma.
