Objective: To explore the security, short-term effect and mechanism ofPVA particles loding nordy perfusion via hepatic artery for rabbit VX2livertumor.Materials and methods: Using the methods of cutting the belly open tomaking the transplanted model of VX2carcinoma in rabbit. Randomlyselected30rabbits VX2liver tumor model, divided into control group(injected with5ml NS), TACE group (injected with0.2ml/kg lipiodol and1mg/kg cisplatin), experimental group (injected with1mg/kg PVA particleswhich combined with nordy), and ten rabbits in each group. Observe thegeneral condition after operation; the changes of Alanine aminotransferase(ALT), aspartate aminotransferase (AST),creatinine (Cr) at1thday before and1,3,7thday after treatment; the blood supply, necrosis and metastasis oftumor tissue were observed by CT before and7,14thday after treatment; themax and minimum diameter of tumor and the tumor volume growth rate; theVEGF at7thday after treatment; rabbits were sacrificed after treatment14thdays, the tumor was fixed with10%formalin, and HE dyeing for observation.Results:1. Basic situation after treatment: After treatment the rabbit in the threegroups had a significantly reduction of sexual activity and listlessness,especially in the TACE group and the experimental group.2. Liver function: The AST and ALT were significant higher at1th day and3thday after treatment in the TACE group and the experimentalgroup, And no significant difference exist between the two groups, butsignificant difference compared with the control group (p=0.000). The twoindexs decreased to the level pre-treatment at7thday after treatment. ButCr was no significant change at1thday and3thday after treatment.3. The image of CT before and after therapy: Most of the tumorswere low-density tumor lesions in the three groups, only a few of tumorswere isodense lesions before therapy.After therapy the tumors existeduneven high-density changes in TACE group and spot-film necrotic foci inexperitamental group, while the tumor low-density crumb of tumor incontrol group.4. The image of DSA after interventional therapy: There were tumornode stain in control group and the ring stain disappeared in experimentalgroup. While there existed significantly iodized oil deposition in TACEgroup.5. Tumor volume growth rate of TACE and experimental group: Thetumor volume growth rate in the two groups were less than control group at14thday after treatment, and the difference is significant(p=0.000).The tumorvolume growth rate in experimental group was less than TACE group.6. The transfer parts in the three groups: liver, lung, lymph nodes, kidney.Tumor metastasis involving all4parts in control group, transfer cases was77.5%(31/40); tumor metastasis involves4parts in TACE group, transfercases accounted for50%(20/40); the experimental group’s tumor metastasisinvolving three parts, transfer cases accounted for only33.3%(10/30).7. The gross morphology for rabbit liver tumor at7thday after therapy.The tumor lesion existed liquefaction necrosis, great tumor cells and invasive growth in control group. While the tumor lesions were isabeling and thereexisted pseudocapsule around the tumor lesions in the TACE group,The tumorcells were degeneration, necrosis and autolysis. the experimental group,compared with the two group, most of the tumor lesions were necrotic fociand new fibrous tissue could be observed.8. VEGF: The VEGF was the highest in TACE group, and there existedsignificant difference compared with the other two groups. This index inexperimental group was higher than control group,and the difference issignificant.Conclusion:1.The treatment for rabbit VX2liver tumor with PVA microspheresloding nordy is safe and short-term effective.2.The growth of tumor could be inhibited by PVA microspheres lodingnordy in some extent.3.the tumor angiogenesis could be inhibited and the tumor metastasiscould be delayed by PVA microspheres loding nordy in some extent.4.PVA microspheres loding nordy provides a new method for thetreatment of tumor.
