Objective: To investigate the protective effects and its mechanismof Fenghuangshan, Pre-and Post-processed Bozhou CortexMoutan on myocardial ischemia reperfusion injury rat.Methods: Fifty male wistar rats were divided into five groups of10animals each by randomization:(1)Sham group;(2)Model group;(3)Fenghuangshan Cortex Moutan group;(4)Pre-processedBozhou Cortex Moutan group;(5)Post-processed Bozhou CortexMoutan group. The myocardial ischemia reperfusion injury modelwas established by a30min occlusion of ananterolateral branch ofthe coronary artery, followed by a60min reperfusion. The Serumlevels of SOD; MDA; GSH; CK; LDH; AST were determined toevaluate the effect of different Cortex Moutan on myocardialischemia reperfusion injury in rat. Determine the infarction area byTTC; To observe the histopathological; The myocardial cellapoptosis was determined by TUNEL method.Results: After60min of reperfusion, a significant decrease in SOD;GSH levels (p<0.01) and an increase in MDA; CK; LDH; AST level(p<0.05) were observed in model group as compared to shamgroup, The model group also increase the infarct size and myocardial cell apoptosis rate induced by myocardial ischemia andreperfusion. Compared with model group, Fenghuangshan; Pre-and Post-processed Bozhou Cortex Moutan could significan-tlydecrease levels of serum MDA; CK; LDH; AST level (p<0.01;p<0.05), improve the content of SOD; GSH levels(p<0.01; p<0.05),and effectively reduce the infarct-ion area and myocardial cellapoptosis rate. However, there are no significant differencesbetween those three groups.Conclusion: Fenghuangshan; Pre-and Post-processed BozhouCortex Moutan exerts protective effects on myocardial ischemiareperfusion injury. The mechanisms might be related to scavengingoxygen free radicals, inhibiting myocardial cell apoptosis, andreducing the myocardial enzymes leaks out. But there are nosignificant difference between those three grops, in other words,those three groups have the same protective effect on myocardialischemia reperfusion injury rat. Therefore, Two factors of differentregions and processing have no influence upon pharmacologicaleffects.
