Objective:Exercise pretreatment can reduce the expression of IL-6 and TNF-? in myocardial ischemia-reperfusion injury of aging rats,so as to reduce the inflammatory response of myocardial ischemia-reperfusion injury,and thus protect the myocardial ischemia-reperfusion injury.Methods:48 healthy male SD old rats were randomly divided into control group(CON),myocardial ischemia-reperfusion injury model group(I/R),exercise pretreatment group+model group(EP+I/R),exercise pretreatment+ control group(EP+ con),12 rats in each group.The model of myocardial ischemia-reperfusion injury(I/R)was established by Langendorff perfusion system.The hearts of con group and EP+ CON group were perfused in vitro for180 min,the hearts of I/R group and EP+ I/R group were perfused in vitro for20 min,then the canning was stopped fo 40 min,and the reperfusion was120 min.During the perfusion period,cardiac function indexes were monitored;at the end of balanced perfusion,5min,30 min,60min and 120 min of reperfusion,coronary outflow fluid was collected to calculate coronary flow(CF,ml/min);TNF-? and IL-6 inflammatory factors in coronary outflow fluid were detected by enzyme-linked immunosorbent assay(Elasa);triphenyl tetrazolium chloride was selected The expression of TNF-? and IL-6 in myocardial tissue was detected by Western blot.Results:1.The effect of EP on the cardiac function of rats in each group:compared with the con group,each index of cardiac function in the I/R group was significantly reduced,and the cardiac function was weakened due to ischemia(P <0.05);compared with the I/R group,EP+ I/R group intervened on the basis of I/R,the index of cardiac function was significantly increased,the function was improved,and the difference was statistically significant(P<0.05);compared with the con group,The indexes of heartfunction in EP+CON group were better than those in con group,the difference was statistically significant(P <0.05).2.The effect of EP on myocardial infarction area of rats in each group:there was no ischemia model in con group and EP+CON group,but no myocardial infarction;compared with I/R group,the infarct area of EP+I/R group decreased significantly,the difference was statistically significant(P<0.05).3.The effect of EP on the content of TNF-? and IL-6 inflammatory factors in the coronary outflow fluid: the expression of TNF-? and IL-6 in con group was at the basic level;compared with con group,the expression of TNF-?and IL-6 in I/R group increased significantly(P <0.05);compared with I/R group,the expression of TNF-?and IL-6 in EP+I/R group decreased significantly(P <0.05).4.The effect of EP on the expression of TNF-?and IL-6 in myocardial tissue:the expression of TNF-?and IL-6 in con group was at the basic level;compared with con group,the expression of TNF-?and IL-6 in I/R group increased significantly(P <0.05);compared with I/R group,the expression of TNF-?and IL-6 in EP+I/R group decreased significantly(P <0.05).Conclusion:1.Exercise pretreatment can induce myocardial ischemia tolerance,reduce myocardial infarction area and improve cardiac function in aged rats with myocardial ischemia-reperfusion.2.Exercise pretreatment can reduce the expression of IL-6 and TNF-? in the myocardial ischemia-reperfusion of the aged rats,reduce the occurrence of inflammatory reaction,and protect the ischemic myocardium. |
