1. Experiment one: The Lipid-decreasing Activity Screening ofDifferent Component Combination of Catsia tora, Salviamiltiorrhiza,Ilex kudingcha and Gynostemma pentaphyllumObjective: To study the effects of different combination of Catsiatora Linn,Salvia miltiorrhiza Bge,Ilex kudingcha C.J.Tseng andGynostemma pentaphyllum (Thunb.)Mak, and screen the optimumcomposite from Composite A (combination of Ilex kudingchaC.J.Tseng, Salvia miltiorrhiza Bge and Catsia tora Linn),Composite B (combination of Gynostemma pentaphyllum (Thunb.) Mak,Salvia miltiorrhiza Bge and Catsia tora Linn), Composite C(combination of Ilex kudingcha C.J.Tseng,Gynostemma pentaphyllum(Thunb.) Mak and Catsia tora Linn) and the water extract of eachsingle traditional medicine herb(Ilex kudingcha C.J.Tseng,Salvia miltiorrhiza Bge, Gynostemma pentaphyllum (Thunb.) Mak andCatsia tora Linn) on down-regulated the serum lipids levels ofhyperlipidemia mice. Futhremore,to establish experimental basement for the further side-effect and mechanism study ofcomponent combination of Ilex kudingcha C.J.Tseng, Salviamiltiorrhiza Bge, Gynostemma pentaphyllum (Thunb.) Mak and Catsiatora Linn.Methods: High fat diet (10%egg yolk,0.2%bile salt,78.8%flour,1%cholesterol,10%lard oil) was used to establish ahyperlipidemia mice model, all the mice were randomly divided intohigh-fat control group (Md), positive control group (Ps), normalcontrol group (Nm),composite A group (C1), composite B group (C2),composite C group (C3), Ilex kuingcha C.J.Tseng group (Cd),Salvia miltiorrhiza Bge group (Sm), Gynostemma pentaphyllum(Thunb.) Mak group (Gp) and Catsia tora Linn group (Ct). The normalcontrol group and high-fat group were given corresponding volumedistill water (20mL·kg-1), and the positive control group was givenSimvastatin tablets(7mg·kg-1). The administer dose of CompositeA was equal to230mg·kg-1Ilex kuingcha C.J.Tseng,197mg·kg-1Salvia miltiorrhiza Bge and1596mg·kg-1Catsia tora Linn, theadminister dose of Composite B was equal to648mg·kg-1Gynostemmapentaphyllum (Thunb.) Mak),197mg·kg-1Salvia miltiorrhiza Bgeand1596mg·kg-1Catsia tora Linn, and the administer dose ofComposite C was equal to230mg·kg-1Ilex kuingcha C.J.Tseng,648mg·kg-1Gynostemma pentaphyllum (Thunb.) Mak) and1596mg·kg-1Catsiatora Linn. Finally, the administer dose of Ilex kuingchaC.J.Tseng group, Salvia miltiorrhiza Bge, Gynostemmapentaphyllum (Thunb.) Mak), and Catsia tora Linn group was equalto1150mg·kg-1Ilex kuingcha C.J.Tseng,985mg·kg-1Salviamiltiorrhiza Bge,3240mg·kg-1Gynostemma pentaphyllum (Thunb.)Mak) and2660mg· kg-1Catsia tora Linn respectively. Thencontinuous administration for4weeks, determinate serum TC, TG and HDL-C of hyperlipidemia mice, as well as body weight and thepathological changes in organs were observed. Results: After4weeks’ consecutive administration, compared with hyperlipidemiamice in model group, serum levels of TC, TG in three compositegroups were significantly lower, as well as the levels of TC andTG in liver, and serum levels of HDL-C was significantly increased,and Composite A and B had stronger effects on reducing the levelsof serum TC, TG and increasing HDL-C than Composite C. Conclusion:Composite A, Composite B and Composite C has remarkable effect onreducing the levels of serum lipid,but Composite A and B arestronger than Composite C.2. Experiment two: Study on the mechanism for the hypolipidemiceffect of Different Component Combination of Catsia tora,Salviamiltiorrhiza, Ilex kudingcha and Gynostemma pentaphyllumObjective: To explore the mechanism for the hypolipidemic effectof Composite A (combination of Ilex kudingcha C.J.Tseng, Salviamiltiorrhiza Bge and Catsia tora Linn), Composite B (combinationof Gynostemma pentaphyllum (Thunb.) Mak, Salvia miltiorrhiza Bgeand Catsia tora Linn)on hypolipidemic.Methods: High fat diet wasused to establish a hyperlipidemia mice model, all the male Kunmingmice were randomly divided into hyperlipidemia model group (Md),positive control group (Ps), normal control group (Nm), CompositeA group (C1), Composite B group (C2)and normal control group(Nm).Then, after continuous administration for4weeks, determinatehyperlipidemia mice in serum levels of TC, TG, HDL-C, SOD (speroxidedismutase), MDA(malondialdehyde), ALT(glutamic pyruvietransaminase), AST(glutamic oxaloacetic transaminase) and NEFA(nonestesterified fatty acid), as well as body weight and observe the pathological changes in liver and other organs, and the biopsystained with Hematoxylin-eosin were observed. Results: Componentcombination of Ilex kudingcha C.J.Tseng, Salvia miltiorrhizaBge and Catsia tora Linn group had a good effect on lowering thelipid. Compared with hyperlipidemic mice in model group, serumlevels of TC, TG in combination of Ilex kudingcha C.J.Tseng, Salviamiltiorrhiza Bge and Catsia tora Linn group were significantlylower, as well as the levels of TC and TG in liver, serum levelsof LDL-C, MDA, ALT and AST; and the concentration of serum HDL-Cand NEFA, the activity of SOD were remarkably increase. Conclusion:Composite A (combination of Ilex kuingcha C.J.Tseng, Salviamiltiorrhiza Bge, Catsia tora Linn) has a good effect on loweringhypolipidemic effect, and its hypolipidemic effect may be relatedto the effect of combination drugs that inhibited the synthesis ofcholesterol and promoted cholesterol metabolism in the liver,improving the body’s antioxidant capacity and coordination theorgan functions of kidney, spleen, liver and protective effect ofliver.
