ObjectiveDetection the expression of nuclear transcription factor-κB and hypoxia-inducible factor-1α in prostate cancer tissue, to explore the relationship between both and prostate cancer’s Gleason score, clinical stage, age. To study whether there is correlation between expression of nuclear transcription factor-κB and hypoxia-inducible factor-1α in prostate cancer.MethodRT-PCR were used to detect NF-κB and HIF-la mRNA expression of12cases of prostate cancer tissue and10cases of benign prostatic hyperplasia tissue, using immunohistochemistry SP method to detect NF-κB and HIF-1α protein expression of50cases of prostate cancer tissue and10cases of benign prostatic hyperplasia tissue.Collected experimental data, used SPSS17.0statistical software for statistical analysis.ResultsRT-PCR results show:the amount of NF-κB mRNA expression in12cases of prostate cancer were0.668±0.045,and the HIF-1α mRNA expression were0.880±0.057;NF-κB mRNA expression level of10patients benign prostatic hyperplasia tissue were0.473±0.032,the expression of HIF-la mRNA were0.586±0.058, relative expression of NF-κB, HIF-1α mRNA in prostate cancer tissue were significantly higher than the benign prostatic hyperplasia group(P<0.05).Immuno-histochemistry SP method results:①the positive expression rate of NF-κB protein in prostate cancer and benign prostatic hyperplasia were62.90%and11.76%, using group chi-square test, and the difference was statistically significant (P<0.05); HIF-1α protein positive expression rate of prostate cancer and benign prostatic hyperplasia group were66.13%and5.88%, using the chi-square test between groups, there are significant differences(P<0.05).②Correlation analysis of NF-κB and HIF-1α protein expression in Prostate cancer tissue with clinical and pathological data shows:patients age≥65years of age, NF-κB protein expression rate were72.73%, patients age <65years old, NF-κB protein expression were51.72%,used chi-square test, the difference had no statistically significant (P>0.05);Same results showed that there was no marked significant of difference between HIF-la protein expression and age (P>0.05). NF-κB protein expression levels of middle-differentiated and poorly differentiated group were significantly higher than that of well-differentiated group in Prostate cancer (P<0.05), HIF-1α protein expression levels of middle-differentiated and poorly differentiated group were significantly higher than that of well-differentiated group in Prostate cancer (P<0.05), Prostate cancer tissue clinical grading were accordance with TMN standardⅢ, Ⅳ stage expression levels of NF-κB protein were significantly higher than those in stage Ⅰ+Ⅱ, which had a significant difference (P<0.05), expression levels of HIF-la prote in Ⅲ, Ⅳstage were significantly higher than Ⅰ+Ⅱ period, which had a marked difference (P<0.05).③NF-κB and HIF-la protein expression analysis results in prostate cancer tissue suggest that:NF-κB and HIF-1α expression in the prostate cancer group was positively correlated (ys=0.368, P<0.05)ConclusionNF-κB and HIF-1α mRNA and protein expression in prostate cancer tissues were significantly higher than the benign prostatic hyperplasia tissues, the positive expression of NF-κB and HIF-1α protein in prostate cancer were gradually increased with the rises of Gleason score and clinical stage. NF-κB in prostate cancer increased expression levels of HIF-1α expression of increased positive correlation.which initially proved that NF-κB activated PI3K/AKt to raise the HIF-1α expression to promote the occurrence, development, invasion and metastasis of prostate cancer.
