Background and ObjectivesGlioma is one of the most common malignant tumors of central nervous system.lt grows invasivly and has no clear boundary with the surrounding brain tissues.Because of difficulty to completely resect,it has high postoperative recurrency, high disablity rate and high mortality.The progrosis of glioma is poor,though the current comprehensive treatment include surgical resection,postoperative radiotherapy and chemotherapy.The therapy of glioma is still one of the most important difficult problems for the neurosurgeon.In recent years,the development of neural molecular biology provides new ways to the study of the biological feature of glioma.Researchers have shown that there are varieties of gene mutations and abnormal expressions in the formation and development of glioma, including proto-oncogenes and tumor suppressor genes and genes related to cell cycle regulation,etc. So the further research of the aforesaid genes in the occurrence and development of glioma as well as the search of the target gene is one of important issues of current neurosurgery.Microtubules has comprehensive biological functions and distinctive cellular dynamics,and play an important role in the chromosome movement and the process of mitosis.α-Tubulin gene encodes a-tubulin,which participates in the formation of microtubules,centrosomes and other important cellular components.The high expression of a-tubulin was found not only in prostatic cancer, breast cancer and lung cancer,but also in precancerous lesions.But there are few reports about relationship between expression of a-tubulin and human glioma.STK15is a cancer gene closely related to the centrosome abnormalities discovered in recent years.STK15is involved in regulation of centrosome maturation, transformation of G2/M phase, establishment of bipolar spindle,chromosome separation,as well as cell division, and is in the strict regulation of time and space.Researchs show that STK15has gene amplification and high protein expression in a wide variety of tumors,and is related to abnormal amplification of cells centrosome and the cell malignant transformation.The abnormal expression of a-tubulin and STK15are closely associated with the occurance and biological behavior of multiple tumors.So we utilize the RT-PCR technique to detect the mRNA expression of a-tubulin and STK15in glioma specimens and investigate their biological significance of expression in the glioma.This may provide the basis for targeted and individualized therapy of glioma.Materials and MethodsWe collected confirmed45glioma samples from Department of Neurosurgery of the Second Affiliated Hospital of Zhengzhou University from July in2011to June in2012,including25males and20females(mean age45.63±15.06).All specimens were obtained from glioma tissues,not including the necrosis of central areas or tissues of tumor border area.All the cases were on the first visit without preoperative radiotherapy or chemotherapy.All the specimens were divided into four grades according to2007WHO Classification of Tumours of the Central Nervous System,including3cases of grade Ⅰ,18cases of grade Ⅱ,16cases in grade Ⅲ,8cases in grade Ⅳ.21cases of grades Ⅰ~Ⅱ were classificated as low-grade group,and24cases of grades Ⅲ~Ⅳ were classificated as high-grade group.12non-tumor tissues were adopted as the control group.All specimens were carefully preservated in-80℃deep cryogenic refrigerator.we detected the mRNA expression of a-tubulin,STK15by RT-PCR in glioma and non-tumor brain tissues.The results were analyzed by the statistics software of SPSS17.0and Probability values a=0.05were considered as statistically significant. Results1. The mRNA expression of a-tubulinThe mRNA expression of a-tubulin in Ⅰ~Ⅱ low grade group was0.905±0.047,and0.976±0.044in Ⅲ~Ⅳhigh grade group,0.943±0.058in the glioma tissues, as well as0.367±0.028in non-tumor brain tissues.There was significant difference between glioma and the non-tumor brain tissues,between high grade group and the non-tumor brain tissues,between high grade group and low grade group,as well as between low grade group and the non-tumor brain tissues in the mRNA expression of a-tubulin (P<0.05).2. The mRNA espression of STK15The mRNA expression of STK15in Ⅰ~Ⅱ low grade group was0.713±0.0210,and0.779±0.018in Ⅲ~Ⅳhigh grade group,0.748±0.038in the glioma tissues, as well as0.455±0.020in non-tumor brain tissues.There was significant difference between glioma and the non-tumor brain tissues,between high grade group and the non-tumor brain tissues, between high grade group and low grade group,as well as between low grade group and the non-tumor brain tissues in The mRNA espression of STK15(P<0.05).3. The correlation between a-tubulin and STK15Through statistical analysis the mRNA expression of a-tubulin show positive correlations with STK15,the Pearson product moment correlation coefficient r=0.754, and it was satistically significant (P<0.05).Conclusions1. The mRNA expression of a-tubulin and STK15in glioma tissue were both significantly higher than in the non-tumor brain tissues,and the expressions increased according to the upgrade of pathological grade of glioma.2. The expression of α-tubulin in glioma shows a significant positive correlation with STK15.
