Background: Breast cancer is one of common malignancies in female. Along with understanding to pathogenesis of the breast cancer , more and more genetic research confirmed that Bag-1,Bcl-2,STK15 and VEGF were correlated with the Carcinogenesis, metastasis and recurrence of breast cancer. The mechanism of these genes and the relationship among these genes and the clinical diagnosis , prognosis of breast cancer were deeply studied, which can provide evidences for clinical diagnosis and treat.Objective: To investigate the expression level of Bag-1, Bcl-2, STK15, VEGF in the benign and malignant breast tumors, and its relationship with clinical predictive indicators.Method: The S-P immunohistochemical method was used to detected the expression of Bag-1, Bcl-2, STK15, VEGF in 89 cases of breast cancer and 20 cases of breast benign lesion, and x~2 test, fisher exact probability method and the spearman correlation were used for statistical analysis.Results: 1 . VEGF positive expression rate in breast cancer was 76.40% , significantly higher than that of benign lesion (p <0.01). The positivity expression of VEGF in breast cancer was correlated with lymph node metastasis and histology stage(P<0.01), but not with the age,tumor size,clinical stage ,ER,PR,HER2. 2. Bag-1 positive expression in breast cancer were 96.63%, significantly higher than that of benign lesions (p <0.01). The positivity expression of Bag-1 had no significant correlation with the patient's age, tumor size, lymph node metastasis ,histological stage ,ER,PR and HER2 receptor status. 3. Bcl-2 positive expression rate in breast cancer were 86.52%, significantly higher than that of benign lesions (p <0.01). The positivity expression of Bcl-2 was correlated with the lymph node metastasis,histological stage(P<0.05), but not with the patient's age, tumor size, clinical stage, ER,PR and HER2. 4. STK15 positive expression rate in breast cancer were 89.89%, significantly higher than that of benign lesions (p <0.01). The positivity expression of STK15 was correlated with the histological grading and HER2(P<0.05), but no with the patient's age, tumor size, lymph node metastasis ,ER,PR .5. STK15,VEGF,Bcl-2 had positive correlation each other;Bag-l had positive correalation with VEGF,but no with Bcl-2 and STK15.Conclusion: The over-expression of Bcl-2,STK15 and VEGF were significantly correlated with the carcinogenesis,development, metastasis of breast cancer. The over-expression of Bag-1 was correlated with the Carcinogenesis of breast cancer. And they had synergetic effection with each other. Bcl-2,STK15 and VEGF may be used as a marker to evaluate the prognosis of breast cancer.
