Objective: It had been found that there is a disorder of Adiponectin leveland abnormal expression of Adiponectin Receptor1(AdipoR1) andAdiponectin Receptor2(AdipoR2) genes in gout, which infer the Adiponectinsignaling pathway may be involved in the pathogenesis of gout. Our project isthat detected the changes of IL-1β concentration after cultivated peripheralblood of the gout patients in which the cell Adiponectin Receptors (AdipoRs)had beed blocked with the anti-AdipoR antibody to explore whetherAdiponectin signaling pathway involved in the gouty inflammation mediatedby IL-1β or not.Methods:20primary acute gouty arthritis patients and20healthyindividuals were enrolled into the study. And the peripheral blood treated withanti-human Adiponectin Receptor1antibody, anti-human AdiponectinReceptor2antibody, anti-rabbit IgG antibody (as negative control) and NS (asblank control) and plasma for detecting IL-1β level by ELISA was separatedfrom blood after cultured with different concentration of anti-AdipoRantibody and time, the results were compared in each group treated withdifferent antibody and time.Results:1.No significant difference was observed between each grouptreated with different concentration of anti-AdipoR1antibody and differentcultured time in both gout patients and healthy controls (P>0.05).2. There aresignificant difference for plasma IL-1β levels in gout patients between groupsof Anti-AdipoR2antibody concentration at1.0μg/ml,2.0μg/ml,4.0μg/ml andblank control(P<0.05, respectively), the dose-effect relationship was observed between the concentration of anti-AdipoR2antibody and the plasma IL-1βlevel. But no significant difference was observed between differentconcentration of anti-AdipoR2antibody in healthy controls(P>0.05). Andthere was no significant for plasma IL-1β level with different cultured time inboth gout patients and healthy controls treated with anti-AdipoR2antibody(P>0.05). There was significant difference in gout patients betweenanti-AdipoR2antibody group and blank control in sametime group(8h、12h、24h)(P<0.05).Conclusion: Adiponectin with AdipoR2signaling pathway may beinvolved in gouty inflammation mediated by IL-1β.
