Dicranopteris Linearis is a kind of medicinal ferns,which has a strong physiological activity, and are widely distributed. Flavonoids is one of the main active ingredient in Dicranopteris Linearis, we deeply researched the extraction, purification, separation, antioxidant capacity and anticancer activity of flavonoids from Dicranopteris Linearis,which provided a theoretical basis for the full develop of Dicranopteris Linearis.(1) In this study, L9(34) orthogonal design Was used in the research of extraction technology of flavonoids from Dicranopteris Linearis. The results showed that:in which ultrasonic was used before microwave was used, the rate of extraction of flavonoids from Dicranopteris Linearis can reached14.47%. The optimum extraction conditions were as follows:material-litjuid ratio1:50, ultrasonic power500W, ultrasonic treatment6min, microwave power700W. microwave pressure0.3Mpa, microwave treatment for2min. Compared to the single microwave method, the extraction rate increased by9%;Ccompared to the single ultrasonic method, the extraction rate increased by15%;Compared to control group, the extraction rate increased by35%.(2) Purified and separated the flavonoids from Dicranopteris Linearis. Flavonoids from Dicranopteris Linearis extracted with95%alcohol, washed with ether-acetone solution, petroleum ether impurity, and then purified by macroporous resin, to extract and seperate different components, we used chloroform, ethyl acetate, butanol, for further separation of the ethyl acetate layer. We took use of gel column chromatography and silica column chromatography Separating the ethyl acetate layer.The experiments show that the purity of purified total flavonoids from Dicranopteris Linearis changed from33.16%to49.48%. For each of the extracted layer, the weight of butanol layer is the biggest, accounting for60.44%, followed by ethyl acetate layer, accounting for21.01%.the chloroform layer was only1.63%. While flavonoids purity of each of the extracted layer size followed by ethyl acetate extraction (75.42%),butanol extraction (55.17%),chloroform extraction (19.04%). The flavonoids of Dicranopteris Linearis are mostly concentrated in the ethyl acetate layer and butanol layer. We get7mian fractions after seperation,the largest component mass fraction in the SU-(1-7) is the component SU-1(81.67%), and it has the highest flavonoids purity, which was77.01%.(3) Through the analysis of the free radical scavenging effects, studied the antioxidant activity of flavonoids from Dicranopteris Linearis. The results show that, after purification, the antioxidant activity of total flavonoids from Dicranopteris Linearis is stronger than that before purification,which indicates the main antioxidant substances is flavonoids from from Dicranopteris Linearis; Among each extraction layer, the ethyl acetate layer has the best antioxidant activity, especially points out the· OH scavenging ability was superior than that of Vc. The IC50value is lower than0.1mg/mL, and when the concentration was0.4mg/mL, The removal rate reached94.55%. Among the seven main groups separated from the ethyl acetate phase, the component of SU-1has the best antioxidant activity, when the concentration was0.2mg/mL, it’s superoxide anion scavenging rate was82.93%.
