Sambucus williamsii Hance leaves contain natural flavonoid,has significant antioxidant and sterilization with equivalent are also found have antitumor activity.This topic research the Sambucus williamsii Hance leaves flavonoids extraction and process optimization,the different polar solvent extracts extraction flavonoids from the effective components and antioxidant capacity were analyzed and discussed and polyamide resin on the extraction of ethyl acetate extract of crude flavonoids were isolated and purified.(1)On the basis of single factor test,response surface method is adopted to optimize the extraction process with ultrasonic assistance for total flavonoids of Sambucus williamsii Hance leaves.Result shows that,the best process condition of extracting total flavonoids of elderberry leaves with ultrasonic assistance is as follows: Ethanol concentration: 72%;Ratio of material to liquid: 42:1;Extracting temperature: 71 ?;Extracting time: 61 min.Under this condition,the highest extraction rate of total flavonoids can reach up to 18.42%.(2)Four kinds of different polar extracts of ethyl acetate,chloroform,butanol and water were extracted by using 3 kinds of different polar solvents.Application of anthrone sulfuric acid method,Shi Atetsunori,vanillin glacial acetic acid perchloric acid method on the relative content of different polarity fractions,polysaccharides,polyphenols,saponins of three effective components determination,and with the obtained flavone content were compared.The flavonoid content of effective ingredients of the extracts of polarity is the highest,and the content of flavonoids in ethyl acetate extract is the highest.(3)antioxidant activity of four different polar extracts(ethyl ether,chloroform,butanol and water)were evaluated by DPPH free radical,ABTS radical,superoxide anion 3 kinds of method.The four polarity extraction constituents of Sambucus williamsii Hance leaves,ethyl acetate,chloroform,butanol and water,have a certain of scavenging activity against DPPH and ABTS free radicals.In fact,the scavenging activity of ethyl acetate is highest,which indicates that,the extraction of ethyl acetate is the optimized part to separate natural antioxidant from Sambucus williamsii Hance leaves.(4)The ethyl acetate extract had the highest flavonoid content,and the antioxidant activity was the highest,so the crude flavonoids were separated and purified.Through the study of static adsorption,desorption,X-5,D101,AB-8,polyamide,silica gel,D-4020 six macroporous adsorption resins were screened,the polyamide resin adsorption rate was 63.31%,the desorption rate is 57.24%,has good ability of adsorption and desorption of flavonoid of ethyl acetate;40% ethanol elution of flavonoids extracts of ethyl acetate in the best purification effect,the relative content of flavonoids 33.1% respectively;elution of ethyl acetate extract of flavonoids of different quality samples with 40% ethanol and the sample was 40 mg,flavonoid compound was purified by polyamide resin purification enriched relative content is the highest;this further purification process conditions and expand the optimization,compared three different types of chromatography columns with different concentration enrichment of flavonoids are relatively content to determine purified by the optimized conditions is available for industrial mass production.
