Commelina communis (Commelina communis L.) is a plant of the genus Commelina communis Commelinaceae, distributed all over the country, mainly from the wild grass. The function of it is water swelling, clearing away heat and purging fire detoxification, clinical mainly for cold and fever, thirst, sore throat, in addition to lowering blood glucose, protecting of liver, antioxidant, antibacterial and other effects.Firstly, The effects of different concentration of ethanol, solid-liquid ratio, ultrasonic time, and temperature on the content of total flavonoids from C.commumnis were investigated by single factor test. Using the content of total flavonoids as index, the extraction technology of C.commumnis was optimized L9 (34) orthogonal experiment and validated on the basis of the results of single factor test, and then compared with the traditional alcohol reflux method (2h). The optimal condition was as follows:25-fold 70% ethanol, extracting for 30min at 70℃.The content of total flavonoids byultrasonic-assisted extraction in Commelina communis L.was 52.51mg/g, while the content of total flavonoids extracted by traditional alcohol refluxing method was 32.34mg/g. The ultrasonic assisted extraction in the extraction time and effects is obviously superior to the traditional alcohol refluxing method, which is suitable for industrialized production.For further purification of the total flavonoids of Commelina communis, we screen these three kinds of macroporous resins (D101, NKA-9, AB-8) by the static saturated adsorption capacity and desorption rate, then, comprehensive compared of the adsorption and separation performance of these three kinds of macroporous resins for total flavonoids of Commelina communis by the recovery rate, and then use of L9 (34) orthogonal experimental design, the establishment of the optimal purification condition of total flavonoids. From the show, AB-8 resin is better than D101and NKA-9 resin. The saturated adsorption capacity of AB-8 resin for total flavonoids of Commelina communis is 32.22 mg/g, desorption rate is 89.53%. After experimental study, optimal purification condition of total flavonoids from AB-8 resin were as follows:the concentration of 1.482mg/ml, pH was 4, the volume fraction of 70% ethanol eluent, elution flow rate of 2 ml/min. The adsorption and separation of AB-8 resin, the purity of the total flavonoids was 63.51%.Furhter, study on the antioxidant activity of the total flavonoids, The antioxidant activity of the extract was tested by using methods of DPPH, pyrogallol autoxidation and phenan throline with Vc as reference. Total flavonoids of Commelina communis compounds have antioxidant capacity, with a dose effect relationship. The scavenging effect of Commelina communis DPPH radical and hydroxyl radical was weaker than that of Vc, while the superoxide anion scavenging effect is stronger than that of Vc. The IC50 of antioxidant activity were 0.0170mg/ml, 0.0055mg/ml,1.9327mg/ml, superoxide anion free radical scavenging capacity of the strongest.
