| Objectives To observe the effects of sinomenine(SIN) on the gastric juice, bile juice and pancreatic juice secretion, and to investigate the regulation of SIN on gastrin(GAS), cholecystokinin(CCK) and secretin(SEC) in the blood of rabbit.Materials and methods24of conventional New Zealand white rabbits were equally divided into experimental and control group through the random number table. The gastric, bile and pancreatic juice of experimental rabbits were collected by gastrostomy, gallbladder fundus ostomy with common bile duct ligation and catheterization near to the opening of pancreatic duct, respectively. SIN was injected in the ear vein of experimental rabbits at the dosage of7.5mg/kg, and in the control group, rabbits were injected the equal volume of Normal Saline (NS). The digestive juices mentioned above were collected at30min before injection of SIN or NS, as well as at30,60,90,120and150min after SIN. Blood samples were got via the ear vein at30min before and at30,60min after injection. Blood GAS, CCK and SEC concentrations were detected by radioimmunoassay. The volume, PH, pepsin concentration in gastric juice, the volume, total bile acid, cholesterol, bilirubin[total bilirubin (TB), direct bilirubin (DB) and indirect bilirubin (IB)] concentration in bile juice, and the volume, PH, protein and amylase concentrations in pancreatic juice were surveyed.Results (1) There were no difference in all of surveyed items between the control and experimental group before the intervention. Compared with control group, the significant increases were found in GAS, CCK and SEC in plasma (at30,60min, p<0.01), the volume of gastric juice (at30min, p<0.05), the volume of bile juice (at30,60,90and120min, p<0.01),the total bile acid concentration (at30,60and90min, p<0.05), the volume of pancreatic juice (at30,60,90and120min, p<0.05), PH of pancreatic juice (at30,60and90min, p<0.01) in the experimental group. In contrast, the experimental group showed decreases in PH of gastric juice (at30and60min, p<0.01), the cholesterol concentration (at30,60and90min,p<0.05), the IB concentration in bile juice (at30,60min, p<0.01) and the amylase concentration in pancreatic juice (at30and60min, p<0.05). No significant difference was found in pepsin concentration in gastric juice, TB and DB concentrations in bile juice and the protein concentration in pancreatic juice between the experimental and control group (p>0.05).(2) There was a significant increase at30and60min post-intravenous injection of SIN in the GAS. CCK and SEC concentration of the experimental group (p<0.01).There was a significant increase at30min post-intravenous injection of SIN in the volume (p<0.05) and pepsin concentration (p<0.01) of gastric juice in the experimental group. The PH of gastric juice decreased significantly at30,60and90 min post-intravenous injection of SIN in the experimental group (p<0.01).The volume and total bile acid concentration of bile increased obviously at30.60and90min post-intravenous injection of SIN in the experimental group(p<0.01). Similarly, cholesterol concentration in the bile was greater at30,60,90and120min post-intravenous injection of SIN in the experimental group (p<0.01). There was no significant statistic difference in the concentration of total and indirect bilirubin in the bile at30,60,90,120and150min post-intravenous injection of SIN in the experimental group (p>0.01), and an obvious increase in the direct bilirubin concentration in the bile was showed at30,60,90,120and150min post-intravenous injection of SIN in the experimental group(p<0.01).The volume of pancreatic juice showed a significant increase at30min post-intravenous injection of SIN in the experimental group (p<0.01). The PH of pancreatic juice increased at30,60and90min post-intravenous injection of SIN in the experimental group (p<0.05). There was an obvious decrease of the protein concentration in pancreatic juice at60,120and150min post-intravenous injection of SIN in the experimental group(p<0.05), while an obvious decrease of the amylase concentration in pancreatic juice at30,60and90min was showed post-intravenous injection of SIN in the experimental group (p<0.01).Conclusions SIN could stimulate the production of gastric, bile and pancreatic juice, along with a syntropic regulation of GAS, CCK and SEC in blood of the New Zealand white rabbits. |
