Research backgroundGlioma is the most common and aggressive primary brain tumor and themechanism of glioma is polygenic disorders. Glioma is the first cancerstudied by The Cancer Genome Atlas (TCGA) and according topathologic stage, the glioma patients must be systematic andpostoperative adjuvant chemotherapy except grade I. Although today themethods of chemotherapy has great progress,only a few patients haveextended survival time. Studies show that the glioma cell have highlyinvasive and easily recurrent, the average survival time of glioma patientshave only9-12months. Therefore, the new anti-tumor chemotherapydrugs which can induce tumor cell apoptosis and inhibit glioma cellproliferation is become a research hotspot.Wogonin is a flavonoid derived from the root of Labiatae skullcap andisolated as a monomer drugs. Wogonin is a widely used Chinese herbalmedicine, which has used in anti-inflammatory therapy. The relativemolecular mass of wogonin is284.26, and its’ molecular formula isC16H12O5. Many studies show that wogonin can inhibit the proliferationof a variety of tumor cells, affect cell cycle and cell apoptosis. And thepossible mechanisms were also researched. The result showed that treatment of U87cells with wogonin induce the cell apoptosis, inhibitedthe cell growth and proliferation in both time-and concentration-dependent manner. The western blot analysis suggested wogonin regulatethe expression of COX-2in human glioma cell. Additionly, we foundthat wogonin-treatment induce the repression of microRNA-128expression affectively and microRNA-128combined with wogonin cangrow the apoptosis and inhibit the proliferation of glioma cell. Overall,these results show that wogonin induce apoptosis through up theregulation of COX-2and miocroRNA-128expression in huamn gliomacell line U87. We believe that wogonin could be a anti-tumor drug infuture.
