Breast cancer is one of the most common malignant tumors of women, the molecular pathogenesis of breast cancer and biological treatment has been the focus of research scholars. Recent studies show that insulin-like growth factor -1 (IGF-1) and its receptor (IGF-1R) play a key biological activity in the cancer cells. Wogonin is isolated in the Scutellaria root,it can block tumor cell signal transduction.It is a potential anticancer drug. The main purpose of this paper is to study the in vitro anti-cancer effects of Wogonin on IGF-1 system in MCF-7 cells. The following are parts of the study:1 The vitro anti-breast cancer effect of wogoninMTT was used to examine the effect of wogonin on MCF-7 cells. Flow cytometry was used to determine the cell cycle distribution.By detecting the supernatant lactate dehydro genase release rate to study cytotoxicity. Application of the appropriate test kit on the cell mitochondrial membrane potential and caspase-3 activity.The results showed that:①Wogonin inhibites the cell proliferation in a dose-and time-dependent.②Wogonin can arrest cell cycle in G0/G1.③Wogonin can increase mitochondrial membrane permeability,caspase-3 activity increased 259.62 percent.2 The in vitro anti-cancer effects of wogonin on IGF-1 system in MCF-7 cellsMTT assay was used to detect the effect of Wogonin on cell viability. Cell cycle distribution and cell apoptotic rate were assessed by Flow Cytometry.ELISA was applied to determine the protein level of IGF-1 and IGFBP-3.Western blottingting was used to detect the effect of Wogonin on IGF-1 stimulated Akt and Erk1/2 phosphorylation The results showed that:①Wogonin inhibited the IGF-1 in promoting proliferation significantly in dose-dependent significant time.②The apoptotic rate of MCF-7 cells was increased after treatment with wogonin and wogonin may lead to cell cycle arrest of MCF-7 cells at G0/G1 phase.③The secretion of IGF-1 of MCF-7 cells was significantly decreased and IGFBP-3 was increased after treatment with wogonin④Wogonin can promote the level of Akt phosphorylation decreased to 58.3±9.0%(P﹤0.05), but the Erk1 / 2 phosphorylation level of no significant impact, indicating wogonin may mainly through the impact of PI3K pathway to achieve on MCF -7 cell line.
