Objective: Expression level of Th17cells and related cytokines ingastric cancer tissue was detected, and the relationship between Th17cells and related cytokines was explored, the possible regulatorymechanisms in gastric cancer was approached, to provide experimentaland theoretical basis for the new treatment strategy.Method:1The peripheral blood Th17lymphocyte proportion were accountedby flow cytometry, characteristics of distribution of Th17cells in theperipheral blood of patients with gastric cancer.2By real-time fluorescence quantitative PCR, the chemokinesCCL1, CCL2, CCL7, CCL17, CCL20, CCL22, CXCL2, CXCL5, IL-8and IL-17mRNA expression in the. gastric cancer tissues and non-cancertissues, tumor draining lymph nodes and non tumor in regional lymphnodes were detected.3By immunohistochemical staining, vascular endothelial theheterosexual antigen CD34and neutrophil-specific antigen CD66b wreedetected.Result:1Compared to healthy controls, The th17cells in the peripheralblood lymphocyte ratio in patients with gastric cancer was significantlyincreased (1.913±0.702%vs.0.402±0.022%, p<0.05)2The quantitative real-time PCR results showed that: comparedwith non-cancer control organizations, gastric carcinoma chemokines ofCCL2, CCL7, CCL20and CCL22mRNA the expression levels werehigher (P<0.05or P<0.01); while CCL17, CXCL2and CXCL5mRNAexpression levels were no differences. Compared with the non-tumor region lymph nodes, draining lymph nodes of tumor area chemokines ofCCL2, CCL7, CXCL5mRNA the expression levels were higher (P<0.05); while CCL17, CCL20, CCL22and CXCL2mRNA were nodifferences.3The relationship of expression of IL-17mRNA and chemotacticfactors mRNA in gastric cancer tissue and tumor draining lymph nodes,found that: gastric carcinoma IL-17mRNA in expression levels ofchemotactic factor CCL20, CCL22mRNA expression significantlycorrelated (P <0.05); expression levels of IL-17mRNA chemokine CCL2,CCL7mRNA expression level was no significant correlation (P>0.05).Tumor area draining lymph nodes IL-17mRNA expression levels ofCCL2, CCL7mRNA expression level was no significant correlation (P>0.05).4Correlation analysis between gastric cancer tissues of IL-17mRNAexpression in gastric cancer patients with clinical and pathologicalparameters: gastric cancer tissues IL-17mRNA in expression levels inrelevant with TNM stage (P <0.05), with the patient’s gender, age, tumordifferentiation, lymph node metastasis, tumor size, independent of thevascular invasion (P>0.05).5Basis the IL-17mRNA expression median gastric cancer tissuewere divided into two groups, the IL-17high expression and lowexpression of IL-17group, immunohistochemistry results showed thatIL-17expression of CD34positive cells significantly higher than thosewith low expression of IL-17. Similarly, IL-17with high expression ofCD66b-positive cell density was significantly higher than the IL-17lowexpression group.Conclusion:1Th17cells was significantly higher expression in the gastric tissueand Peripheral blood, the Th17cells in microenvironment gathered maybe because of chemokines CCL20, CCL22expression increasedinduction of Th17cells from the peripheral to the tumor tissue. 2The Th17cells aggregation, and promote gastric cancerprogression by inducing angiogenesis and neutrophil infiltration in gastriccarcinoma.
