This dissertation consists of four parts together. Summary of the relevant literatures; Researches on the chemical constituents of (Dioscorea collettii Hook. f. var. hypoglauca(Pslibin)Pei et Ting); Studies on the extraction and purification technology of the saponins fractions; Researches on the quality control of (Dioscorea collettii Hook. f. var. hypoglauca(Pslibin)Pei et Ting)Chapter I Summary of the related literaturesDioscorea hypoglauca Palibin.is the dried rhizome of the Dioscorea collettii Hook. f. var. hypoglauca(Pslibin)Pei et Ting. Its favor is bitter and plain and It belonges to kidney, stomach. In this part, researches on the chemical constituents, pharmacological action and clinical application of Hypoglaucous Collett Yam Rhizome were reviewed on the basis of77references.Chapter II Researches on the chemical constituents of (Dioscorea collettii Hook. f. var. hypoglauca(Pslibin)Pei et Ting)By using several isolation methods and determination technologies including TLC, HPLC,’HNMR,13CNMR, there were18compounds obtained from Dioscorea hypoglauca Palibin. Among these18compounds, the detailed structures of13compounds were indentified so far. They are three diarylheptanoids:tsaokoarylone,1,7-bis(4-hydroxyp henyl)-4,6-heptabien-3-one,1,7-bis(4-hydroxyp henyl)-1,4,6-heptatrien-3-one; two quinones:emodin and7-hydroxy-2,6-dimethoxy-1,4-phenanthrenequinone; two seroids: β-sitosterol and stigmasterol; aliphatic acid:palmitic acid and hexacosanoic acid; one Coumarins:(3S)-6,8-two hydroxy-3-phenyl-3,4-dihydro isocoumarin; one flavonoids: kaempferol; two sapogenins:diosgenin and ruscogenin; the other three:5-hydroxymethylfurfural. Among them,7-hydroxy-2,6-dimethoxy-1,4-phenanthrenequinone, stigmasterol, emodin, hexacosanoic acid, kaempferol and ruscogenin were firstly found from Dioscorea hypoglauca Palibin, meanwhile tsaokoarylone,5-hydroxymethylfurfural were firstly found from Dioscorea.Chapter III Studies on the extraction and purification technology of the saponins fractionsThe extraction technology of saponin fraction of Dioscorea hypoglauca Palibin has been studied. L9(34) orthogonal test has been used to evaluate the best extraction technology, including the best extraction solvent, amount of the solvent, hours and number of times, of saponins extraction which was evaluated by the extraction efficiency of total saponins. After evaluation, the best technology is that using B1BV times dosage A2%ethanol, to extract the raw medicine material D3times, C2hours every time. The extraction technology referred above was tested and verified. The preparation and determination results implied that the established extraction technology was steady and could be repeated.The purification technology of saponins fraction of Dioscorea hypoglauca Palibin has been established. After testing the absorptive property of six different types of macroporous absorption resins, C type was selected because of its preeminent absorptive and separate property of saponins fraction. In order to optimize the purification technology of purifying the saponins fraction of Dioscorea hypoglauca Palibin with C type, such as the concentration of the solution, velocity of absorption, the ratio of diameter and height, loading capacity, purifying solvent, eluting solvent, the amount of purifying and eluting solvent, and et al, we figure out the best purification technology of saponins fraction of Dioscorea hypoglauca Palibin, which is as follow:firstly the extraction fluid was concentrated in I2g/mL, absorbed by C (diameter/height1:G2of resin column) with the velocity of flow of J2BV/h, and the proportion between the total amount of the solution and the resin column was R,3BV of water then L2BVofK2%ethanol to wipe off impurity with P2BV/h, then N2BVofM2%ethanol to elute with S1BV/h. Finally, the extraction and purification technology described above was verified through3batched of samples of saponins fraction of Dioscorea hypoglauca Palibin. The preparation and determination results showed that established extraction and purification technologies were steady and could be repeated. In the saponins fraction, the content of saponins was nearly53%and the transferring ratio was60%, the result was steady and convincible.Chapter Ⅳ Researches on the quality control of Dioscorea hypoglauca Palibin.Perchloric acid colorimetric were established for the determination of the total saponin content of the saponin fraction of the Hypoglaucous Collett Yam Rhizome herbs diosgenin as reference, the measured absorbance at408nm at the color diosgenin standard curve was Y=0.1383Ⅹ-0.1582, r=0.9992, showed that the diosgenin absorbance linearity in the range of2.86~20.05μg/mL is good. The results show that the method is simple, accurate, measured as the Hypoglaucous Collett Yam Rhizome herbs total saponins method.Innovation: 1.18compounds were obtained from Dioscorea hypoglauca Palibin. Among these19compounds, the detailed structures of14compounds were indentified so far. Among them,7-hydroxy-2,6-dimethoxy-1,4-phenanthrenequinone, stigmasterol, emodin, hexacosanoic acid, kaempferol and ruscogenin were firstly found from Dioscorea hypoglauca Palibin, meanwhile tsaokoarylone,5-hydroxymethylfurfural and dibutyl phthalate were firstly found from Dioscorea.2. The purification technique of saponin fraction of Dioscorea hypoglauca Palibin was established for the first time..3. The determination method of saponin fraction of Dioscorea hypoglauca Palibin was established for the first time.
