Irritable Bowel Syndrome (IBS), a usually known functional bowel disorder and a typical psychosomatic disease, is a series of bowel syndromes with characters of choric and repeated abdominal ache or discomfort, and of abnormal defecation. It’s clinical manifestations are abdominal ache, diarrhea, exigency of defecation but incompletion, constipation, alternate diarrhea and defecation, abdominal distention, borborygmus, and exhaust. The pathogenesis is undefined. Research shows that, IBS rats’gastrointestinal hormone is abnormal, and gastrointestinal diseases go with intestinal microcirculation disorder. Acupuncture can both adjust levels of gastrointestinal hormone and have obvious curative effect on IBS. So, this experiment is to EA Zusanli Point, Tianshu Point, to observe effects on IBS rats’ mesenteric microvascular’ diameter, value of CGRP, VIP, and ET, and to have a further discussion about a possible mechanism of IBS’s acu-puncture treatment.Objective:To observe effects of EA Tianshu Point, Zusanli Point on IBS rat models’splanchnic sensitivity, intestinal microcirculation, and changed value of CGRP, VIP, and ET in IBS rat models’plasma, and to discuss parts of the mechanism of IBS’s acu-puncture treatment.Methods:To set WISTAR young rats as the object of the study and to establish models of irritable bowel syndrome rats. The experiment included normal group, model group, Tianshu group and Zusanli group. The Tianshu group and Zusanli group were started to be bounded and given EA in the6th week.once in two days,a total of seven times. Model group was bounded but not given EA, the normal group was done nothing. After the treatment. To use XW-B-3Cold Lightmicro-circulation tester to observe the rats’ state of caps and blood How of mesentery microcirculation in each group and different time period. After the observation, phlebotomize5ml abdominal aortic blood in the situation under anesthesia. Put the blood in EDTA, centrifugalize, and put the supernatant into the refrigerator of-20for measuring. Measure value of plasma CORP. VIP. and ET in rats’ plasma with Elisa.Results:(1) the estimate of the visceral sensitivity:compared with the normal group, the value of the model group rats’ belly raising and back hunch-up significantly reduced (P<0.01). Compared with the model group, the value of the EA Tianshu and Zusanli group rats’ belly raising and back hunch-up significantly increased (P<0.01).(2) test of the mesentery microcirculation:①the diameter of the blood vessel:compared with the normal group, the diameter of the mesentery micro-vessels of the model group significantly narrowed (P<0.05); Compared to the model group, the diameter of the mesentery micro-vessels of the EA Tianshu group and Zusanli group significantly expanded (P<0.05~0.01).②the state of the blood flow:compared with the normal group, the model group’s blood flow speed of mesentery micro-vessels significantly reduced or stopped (P<0.01).(3):Value of CGRP VIP and ET in plasma:compared with the normal group,The content of plasma VIP and ET were significantly ascent in model group,(P<0.01. P<0.05); The content of plasma CGRP and VIP were significantly ascent in EA zusanli group,(P<0.05、 P<0.01). Compared to the model group, the content of plasma CGRP was significantly ascend(P<0.05), while the content of VIP was significantly descend(P<0.01)in EA Tianshu group.The content of plasma CGRP was significantly ascent in EA Zusanli group (P<0.01).Conclusion:1. EA Tianshu, Zusanli can significantly increase the value of the visceral Sensitivity threshold of the IBS rats and reduce the visceral sensitivity.2. EA Tianshu, Zusanli can also significantly improve the blood state of the mesentery microcirculation and remove spasm to relieve the lack of blood and oxygen in the intestinal mucosa. As a result, it has the therapeutic effect of pain relief.3. The levels of values of plasma CGRP, VIP, and ET in irritable bowel syndrome model rats were abnormal, leading to imbalance of these three elements, and this may be one of the mechanisms which cause irritable bowel syndrome.4. Acupuncture can cure irritable bowel syndrome by adjusting the disorder of these three elements.5. EA Zusanli and Tianshu both have characteristic adjustment effects. Above all,abnormal plasma gastrointestinal hormone and intestinal microcirculation disorder are parts of IBS’s mechanisms. A part of EA’s cured effective is to adjust abnormal gastrointestinal hormone, CGRP, VIP, ET, and to have further effects of expanding mesenteric microvascular’ diameter, curing microcirculation disorder, and relief of pain. In addition, very point has its character. Zusanli has a comparatively obvious effect of expanding mesenteric microvascular’diameter.
