Objective Aplastic anemia (AA) is a heterogeneity disease,caused the failure to the bone and in hematopoietic function by a variety of etiology, caused pancytopenia in clinical.The pathogenesis of AA is complex,concerning hematopoietic stem cell damage, dysimmunity, hemopoietic microenvironment change. Plenty of clinical and experimental data show that the main pathogenesis on immune function abnormality, immune disorders and cell factors secreted disorders participated in the incidence of aplastic anemia. Aplastic anemia immune disease process is basically:some unknown factors activate t-cells Th1cells, that number, CD8+cell proliferation, negative hematopoietic regulatory factors IL-2, IFN-γ, TNF-α secrete excessive, triggering Fas/FasL mediated hematopoietic stem cell apoptosis, eventually leading to the haematopoietic function failure. IL-17is the newly discovered a former inflammatory cells factor, mainly by CD4+memory activation T lymphocyte (Th17) produce. Its biological function has been widely studied. At present, in all sorts of autoimmune diseases, including rheumatoid arthritis, multiple sclerosis, systemic lupus erythematosus and asthma patients serum and organization detected IL-17high expression. IL-17by inducing inflammation, chemotactic factors that cause tissue organ such as the inflammatory lesions. Aplastic anemia, as a kind of immune diseases, IL-17in the pathogenesis of aplastic anemia does play a role in the process needs to be explored. This topic respectively by means of ELISA and immunohistochemical method to detect first aplastic anemia patients, treated patients and controls were content of bone marrow fluid and bone marrow tissue IL-17expression, to explore IL-and the role in the pathogenesis of aplastic anemia.Methods Adopt enzyme-linked immunosorbent method (EILSA) detection12cases of aplastic anemia patients (aplastic anemia group),11cases of treatment of aplastic anemia patients (treatment group) and12cases of blood, immune system disease patients (control group) bone marrow in qing express;17IL Adopt immunohistochemical method (IHC) detection26cases in treatment group begin aplastic anemia patients21cases of treatment, after patients (aplastic anemia relief in treatment group) and20cases of blood, immune system disease patients (control group) marrow tissue IL-17expression.Results①IL-17were expressed in aplastic anemia group,treatment group and control group.②The protein content of IL-17in supernate of bone marrow in aplastic anemia group was (35.064±10.857)pg/ml, the positive expression rate of IL-17in Bone marrow tissue was88.5%, the protein content of IL-17(117.678±9.427); the protein content of IL-17in supernate of bone marrow in treatment group was (14.512±2.987)pg/ml, the positive expression rate of IL-17in Bone marrow tissue was71.4%, the protein content of IL-17(143.551±12.311); the protein content of IL-17in supernate of bone marrow in control group was (6.196±12.311)pg/ml, the positive expression rate of IL-17in Bone marrow tissue was40%, the protein content of IL-17(159.658±14.069). The level of IL-17expression in aplastic anemie group was higher than that in treatment group and control group(P<0.05).③The level of IL-17expression in treatment group was higher contro group (P<0.05).Conclusion The expressions of IL-17in AA patients bone marrow tissue is the heightest, the expression of IL-17is reduced after therapy,this hint IL-17may has played a certain role in the pathogenesis of the immune in aplastic anemia.
