Objective:Determination of the length of Telomere(TEL)and the mRNA expression of autophagy related protein P62,Beclin1 and LC3B in bone marrow mononuclear cells of aplastic anemia patients.In order to study the correlation of AA TCM dialectical type and bone marrow mononuclear cells of the lenghth of telomere,and autophagy related protein P62,Beclin1,LC3B mRNA express level,further investigate the relationship between autophagy and the incidence of AA was further explored.Method:79 patients with AA were grouped in accordance with the TCM Syndrome Differentiation standard,"Ji lao sui ku group" 21 cases,"Shen yang xu group" 19 cases,"Shen yin xu group" 19 cases "Shen yin yang lang xu group" 20 cases.And 24 normal human bone marrow mononuclear cells were selected as control group.Using Real-time quantitative PCR method to determine the 79 cases of patients with the length of telomere,autophagy related protein P62,Beclin1,LC3B mRNA expression levels in bone marrow mononuclear cells.Observe the correlation of AA TCM dialectical type and bone marrow mononuclear cells of the length of Telomere,autophagy related protein P62,Beclin1,LC3B mRNA expression levels.To study the correlation between Telomere,P62,Beclin1 and LC3B in bone marrow mononuclear cells of patients with Aplastic Anemia.To study the correlation between telomere length,P62,Beclin1 and LC3B in AA patients' bone marrow mononuclear cell.Study of CAA,SAA patients telomere length(TEL)and P62,Beclin1,LC3B mRNA expression level and peripheral blood CD8+,CD4+ T lymphocyte percentage of correlation.Result: 1.The telomere length of CAA patients and SAA patients was significantly lower than that in the normal control group.And the expression level of P62 mRNA in CAA patients and SAA patients was significantly higher than that in the normal control group.The expression of Beclin 1 mRNA in CAA patients and SAA patients was significantly lower than that in the normal control group.The expression of LC3B mRNA in CAA patients and SAA patients was significantly lower than that in the normal control group.And SAA patients was significantly lower than CAA patients.The above results were statistically significant.2.The expression level of telomere length in AA patients of "Ji lao sui ku group","Shen yin xu group","Shen yin yang lang xu group" were lower than "Shen yang xu group",which have statistical significance.Moreover,the expression level of each group was lower than that of the normal control group.The expression level of P62 in AA patients of "Shen yin xu group","Shen yin yang lang xu group","Ji lao sui ku group" were higher than "Shen yang xu group",which have statistical significance.Moreover,the expression level of each group was higher than that of the normal control group.The expression level of Beclin1?LC3B in AA patients of "Shen yin xu group","Shen yin yang lang xu group","Ji lao sui ku group" were lower than "Shen yang xu group",which have statistical significance.Moreover,the expression level of each group was lower than that of the normal control group.3.Telomere length was negatively correlated with the expression level of P62 mRNA(R=-0.384,P<0.05),and telomere length was positively correlated with the expression level of LC3B mRNA(R=0.315,P<0.05).P62 was negatively correlated with Beclin1 and LC3B mRNA expression levels(R=-0.365 R=-0.913,P<0.05).4.The percentage of peripheral blood T lymphocytes in group CAA was lower than that in group SAA.The percentage of CD8+ in peripheral blood of group CAA was lower than that in group SAA.There was no significant difference in the percentage of CD4+ and CD4+/CD8+ in peripheral blood between group CAA and group SAA.The expression of telomere length(TEL)was negatively correlated with T lymphocyte%,CD8+% and CD4+%;the expression level of P62 mRNA was positively correlated with the percentage of T lymphocyte,CD8+% and CD4+%,and the expression level of Beclin1,LC3B mRNA was negatively correlated with the percentage of lymphocyte.Conclusion:1.Autophagy related proteins P62,Beclin1,LC3B and telomere can participate in the pathogenesis of AA,and the high expression of P62 and low expression of LC3B may be related to telomere length.2.Shortening of telomere length plays an important role in the pathogenesis and progression of AA.The lowest expression was found in "Ji lao sui ku group".3.The expression of P62 increased in the bone marrow of AA patients,and it was the highest in "Ji lao sui ku group",indicating that the accumulation of waste cells and the decrease of autophagy level.Beclin1 and LC3B were low in the bone marrow of AA patients,the lowest in "Ji lao sui ku group",suggesting a decrease in autophagy.4.Telomere length was negatively correlated with the expression level of P62 mRNA,and telomere length was positively correlated with LC3B mRNA expression level.P62 was negatively correlated with Beclin1 and LC3B mRNA expression levels.5.The telomere length(TEL)and P62,Beclin1 and LC3B mRNA expression levels in AA patients were correlated with CD8+,CD4+ and T lymphocytes in peripheral blood.
