Lung cancer is the most common human malignancy tumor. Clinically lung cancer mainly includes non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC), about80%of lung cancer patients belongs to NSCLC. The descent of the lung cancer with transfer is a complex, many steps, many link process, involving the tumor cell proliferation and adhesion, movement, degradation and membrane and the extracellular matrix(ECM), etc. Normal alveolar epithelial basement membrane is constituted by Ⅳ type collagen components, matrix metalloproteinases9(MMP-9) is the major enzymes of matrix metalloproteinases family (MMPS) which degrades Ⅳ type collagen, MMP-9and its specific tissue inhibitors of metalloproteinase1(TIMP-1) plays an important role to attack and the formation process of metastases in tumor cells. At the same time, NSCLC is a typical vascular dependence lesions, the formation of new blood vessels promot the growth of tumors, invasion and metastasis, microvascular denstity (MVD) is the measure of tumor angiogenesis quantitative index, which is a study as a tumor angiogenesis symbols to research and tumor of the relationship. CD105is currently accepted as the more sensitive marker of the blood vessels, which has strong, clear, and dyeing background to count and other advantages. The expression of CD105shows that the tumor microvascular reflect tumor tissue MVD, thus quantitative analysis of tumor microvascular hyperplasia. This study applies immunohistochemical SP method and Western blot method to detect MMP-9and TIMP-1expression in NSCLC and cancer adjacent tissues, application immunohistochemical SP method to detect CD105expression in NSCLC and adjacent tissues of NSCLC to measure MVD, which can understand completely the relevance of NSCLC biological effects and expression and the clinical and pathological features.[Objective](1) To investigate the expression and relationship of tumor biological behavior of matrix metalloproteinases9(MMP-9) and its specific inhibitors tissue metal protease inhibitors-1(TIMP-1) in NSCLC of;(2) To explore the clinicopathologic correlation of CD105expression and microvascular denstity (MVD) detection in NSCLC;(3) To detect the diagnosis and significance of MMP-9, TIMP-1and CD105in treatment of NSCLC, trying to find the value judgment tumor marker NSCLC.[Method]50cases NSCLC wax piece of samples and26patients diagnosed after surgery for NSCLC specimens were collected.(1) The morphology of26cases NSCLC specimens which were diagnosed after surgery were observed.(2) The morphology change and clinical relationship were observed by HE staining in the NSCLC corresponding normal tissue;(3) MMP-9, TIMP-1, CD105expression in76patients with NSCLC and adjacent normal tissue were observed by immunohistochemical SP method, and analyzed the correlation of its expression with lung cancer types, tumor size, TNM staging, differentiation degree, lymph node metastasis.(4) CD105protein expression in26cases of surgical specimens of NSCLC were observed by Western blot;(5) According to the results of immunohistochemical CD105, the relationship between the NSCLC and vessels density through the research of computer image analysis;(6) According to average light density values and Western blot grey value, significance of MMP-9, TIMP-1and CD105in the diagnosis and treatment of NSCLC were discussed by SPSS for Windows13.0statistical software processing image analysis data immunohistochemical.[Results](1)16cases (61.5%) middle of the bronchial type (lung door type) in26cases which were diagnosed after surgery for NSCLC specimen were occured in main and leaves bronchi,9cases (34.6%) peripheral lung cancer originated in the surrounding isolated lung nodules,1case (3.9%)from peripheral lung tissue named diffuse lung cancer;(2) NSCLC histologic appearance was complex with HE staining observation,49cases (64.5%) in76patients of NSCLC was squamous cell carcinoma, with a central lung cancer was the most common, especially older men, and a close relationship with smoking;21cases (27.6%) in76patients with NSCLC was adenocarcinoma, more refered to women, peripheral lung cancer;6cases (7.9%) was large cell anaplastic carcinoma (large cell carcinoma), with high malignant degree, early and extensive transfer, shorter survival period.(3) Positive expression rate of MMP-9, TIMP-1and CD105in lung cancer tissue was significantly higher than that in the normal tissue adjacent carcinoma (P<0.05). MMP-9, TIMP-1and CD105expression in NSCLC were significant correlation with tumor differentiation degree, the clinical stages, lymph node metastasis, distant metastasis (P<0.05or P<0.01).(4) CD105protein expression, according to the results of Western blot,26cases NSCLC organization was significantly higher than those in the corresponding normal tissue adjacent to cancer (P<0.05).(5) Microvessel density (MVD) was correlated with NSCLC differentiation degree, the clinical stages, lymph node metastasis, adenocarcinoma expressed more obviously than squamous cell carcinoma through the computer image analysis.[Conclusion](1) The central type NSCLC (lung hilus type) is primarily in the region, next is peripheral type lung cancer, diffuse type lung cancer is rare.(2) The tissue from the original tumor more than10cm could be used as normal control group; NSCLC histologic appearance shows the squamous cell carcinoma is the most common, adenocarcinoma next, large cell carcinoma is a rare. there is a relationship of histological classification with the prognosis of patients.(3) Positive expression rates of MMP-9, TIMP-1and CD105in lung cancer tissue of are significantly higher than those in the adjacent carcinoma normal tissue and correlation with tumor differentiation degree, the clinical stages, lymph node metastasis, distant metastasis.(4) The proteins expression of CD105in NSCLC are significantly higher than those in the corresponding normal tissue.(5) Microvessel density (MVD) is correlated with tumor differentiation degree, the clinical stages, lymph node metastasis, adenocarcinoma expressed more obviously than squamous cell carcinoma.(6)Detection of MMP-9, TIMP-1and CD105expression and MVD in NSCLC has certain significance to judge the malignant tumor density degree and estimate prognosis.
