Abstract/Summary:
Cardiovascular disease is a severe health menace to human being. Due to its characteristic of integrated effects of multi-component, multi-target and multiple way, TCM gained a great favor of researchers. Based on this reason, drug pairs Carthamus tinctorius L. Caesalpinia Sappan L. with relationship of " Xiangxu " which treated blood stasis syndrome was taken as the research object, the metabolic profiling in normal/model rats administrated with single drug and drug pairs were observed with the aid of HPLC/MS metabonomics method, meanwhile, the relationship of medical components group, metabolites group of medical components and endogenous responsive substances group in plasma was investigated by using a mathematical model of nonstationary random procedure. This study will explicit the substantial base of the drug paris above.1. A high performance liquid chromatographic analysis on HSYA in plasma of rats was developed. According the standard testing of biological samples FDA U.S., the results of methodological evaluation showed it can be successfully used in the research of pharmacokinetics of HSYA.2. Based on the analysis method above, pharmacokinetics of HSYA in normal rats were investigated after administrated Carthamus tinctorius L. and the drug pairs Carthamus tinctorius L.-Caesalpinia Sappan L., respectively. And then, correlative analysis and PCA(Principal Components Analysis) were adopted in the data analysis. The main results are as follows:①Compared the Carthamus tinctorius L group, the half-life of distribution was decreased in rats administrated with the drug pairs; and that indicated Caesalpinia Sappan L. could promote the distribution of HSYA in vivo.②HSYA, ES16、ES35、ES37NM18、NM19is main reason which lead to a significant difference in the two groups from the results of correlative analysis and PCA, perhaps it has a relationship with the substantial base of drug pairs Carthamus tinctorius L.-Caesalpinia Sappan L. in normal rats. 3. The pharmacokinetics of HSYA in stagnancy of Qi and Blood Stasis model rats were investigated after administrated Carthamus tinctorius L. and the drug pairs Carthamus tinctorius L. Caesalpinia Sappan L., respectively. Consequently, correlative analysis and PCA(Principal Components Analysis) are adopted in the research. The main results are as follows:①Compared the Carthamus tinctorius L group, the half-life of distribution, area under the curve and peak concentration of HSYA were increased in model rats administrated with the drug pairs; and that indicated Caesalpinia Sappan L. could slow down the distribution of HSYA and increase the absorption of HSYA in vivo.②HSYA, ES16, ES35, ES51, NM18, NM19, NM23, NM26and NM29is main reason which lead to a significant difference in the two groups from the results of correlative analysis and PCA, maybe it has a relationship with the substantial base of drug pairs Carthamus tinctorius L.-Caesalpinia Sappan L. in the model rats.4. Some effective substance including HSYA, ES16, ES35, ES37, ES51, NM19, NM23, NM26, NM29were analyzed with qualitatively HPLC-MS primarily.
