This dissertation is composed of three chapters. The first chapter summarized the progress of chemical constituent and pharmaceutical activity of Caesalpinia Linn. The second chapter studied chemical compounds from C. sappan L pieces.The third chapter to study the effects of C. sappan L pieces of tyrosinase activity.Chapter 1. Research progress on chemical compounds and pharmacological effects of Caesalpinia L.The progress of chemical constituents and pharmacological research of Caesalpinia L. were summarized.Chapter 2. Study on the chemical constituents of Caesalpinia sappan Linn. piecesUsing silica gel and Sephadex column chromatography, preparative HPLC chromatography and waters high performance liquid chromatography(HPLC), of 70% ethanol extract of C. sappan pieces chemical constituents were isolated and purified, and the use of NMR and MS spectral data to determine the combination of physical and chemical properties compound structure. From the70% ethanol extract of C. sappan pieces dissolved in methanol on macroporous resin column,methanol elution Part 40% methanol and 60% methanol were isolated and identified 9 compounds,respectively(3R,4S)-3-(3',4'-hydroxybenzyl)-3,4-dihydro-2'',3''-dimethyl-3H-[1,3]dioxolo[4,5-c]-chromen-7-ol(1), brazilin(2), protoasppanin A(3), protoasppanin C(4), protoasppanin B(5),isoprotosappanin B(6), caesalpin J(8), sappanone B(9), sappanchalcone(10).Chapter 3. Study on the effect of tyrosinase activityOn tyrosinase inhibitors of kojic acid as a positive control, 70% ethanol extract of C. sappan pieces different fractions and compounds of study on effect of tyrosinase activity in vitro. The results show that in addition to the site of Me OH and other six parts of the C. sappan pieces on tyrosinase activity were activation, which parts of the 40%Me OH on tyrosinase activation and inhibition are weak.Compounds were isolated from the site of 40%Me OH compound(3R,4S)-3-(3',4'-hydroxybenzyl)-3,4-dihydro-2'',3''-dimethyl-3H-[1,3]dioxolo[4,5-c]chromen-7-ol(1), brazilin(2), protoasppanin C(4) and sappanone B(9) isolated from the site of 60%Me OH on tyrosinase activity were activation; compound protoasppanin A(3), CL-7 and caesalpin J(8) on the activity of tyrosinase has inhibitory effect; The isomers of the protoasppanin B(5) and isoprotosappanin B(6) in the mixture of tyrosinase effect no inhibition and no activation.Three compounds on the activity of tyrosinase inhibition type and mechanism research found that the original compounds protoasppanin A(3) and CL-7 on the activity of tyrosinase inhibition type for competitive inhibition, inhibition mechanism is only binds the free enzyme,reduce the binding DOPA and enzyme, so as to inhibit the tyrosine enzyme; compound caesalpin J(8) on tyrosinase inhibition for anti competitive inhibition, inhibition mechanism: inhibitor does not bind to the free enzyme, which only binds the enzyme substrate complex.Taking vitamin C as positive control, using SD rats for 7 days after the drug was detected in the serum levels of tyrosinase, of 70% ethanol extract of C. sappan pieces different groups and some of the compounds were of in vivo tyrosinase activity. The results showed that 70% ethanol extract of C. sappan pieces can reduce the serum content of tyrosinase in rats; compound brazilin,protosappanin A, protosappanin B and caesalpin J promote formation of rat serum tyrosinase, only sappanone B is inhibited the synthesis of tyrosinase in serum of rats.
