Objective: To investigate the influence mechanism of volatile constituents of four resuscitation-inducing aromatic herbs on sodium di-thionote (Na2S2O4)-in duced oxygen glucose deprivation in differentiated rat pheochromocytoma (PC12) cells. From the cellular and molecular level to explain the influence mecha nism and the material basis of the protective effects of neuronal.Methods: oxygen glucose deprivation model of PC12cells was establis-he d by exposure to of Na2S2O4,to study morphological changes in cells, The pr-otective effects of volatile constituents of Moschus, Styrax, Benzoinum extractio n by water-vapor distilling and Borneolum Syntheticum on the model were eva-luated by cytomorphology and colormetric MTT assay. By spectrophotometry, The concentration of intracellu-lar Ca2+、NO、5-HT、DA and NE was assayed by the determination system.Results:①Moschus (0.4,0.8,1.6μg/100μl), Borneolun Syntheticum (0.8、1.6μg/100μl), Styrax (2.0,4.0μg/100μl) and Benzoinum (1.6μg/100μ1) markedly increased the optical density at490nm tested by colorimet-ric MTT assay (P<0.05, P<0.01) and showed their significant effects in protecting PC12cells again st Na2S2O4-induced apoptosis.②Styrax (4.0、8.0μg/100μl) and Benzoinum (0.4、0.8、1.6μg/100μl) markedly att-enuated the concentration of intracellular Ca2+(P <0.05, P<0.01).Moschus and Borneolun Syntheticum showed the tendency of reducing the concentration of intracellular Ca2+.③Bomeolun Syntheticum (0.8、1.6、3.2μg/100μl), Styrax (2.0、4.0、8.0μg/100μl) and Benzoinum (0.8、1.6μg/100μ1) markedly attenuated the concentration of intracellular NO (P<0.05, P<0.01).④Styrax (4.0、8.0μg/100μl) and Benzoinum (1.6μg/100μl) significantly incre ased the concentration of the5-HT(P<0.05, P<0.01); Benzoinum (0.4、0.8、1.6μg/100μl) significantly increased the concentration of the DA(P<0.05, P<0.01); Moschus, Borneolun Syntheticum, Styrax and Benzoinum showed the tende ncy of reducing the concentration of the NE,but has no significantdifference (P>0.05). Conclusion:The volatile constituents of Moschus, Styrax, Benzoinum and Borneolum Syntheticum can markedly protect PC12cells from oxygen glucose deprivation injury through attenuating the concentrat-ion of intracellular Ca2+an d preventing calcium overload, accommodating the turbulence of Monoamine Neurotransmitters. It shows that the volatile constituents of four resuscitation-in ducing aromatic herbs are the material basis of the protective effects of neuron al.
