Objective:The present study was designed to observe the effects of behavior and brainmonoamine neurotransmitters nor epinephrine and5-hydroxyptanmine in chronicunpredictable mild stress depression model rats and to explore its mechanism withantidepressant Ganmai Dazao Decoction(甘麦大枣汤),then compare the effects ofdifferent doses.Methods:1, Model preparation: Choose60weight (180-200g) grams of clean grade SDmale rats were randomly assigned into normal control group, model groupVenlafaxine group, Ganmai Dazao Decoction groups, scattered high, medium andlow does group,10in each group. Normal group every cage5raise,normal diet, nomodel preparation,the remaining50preparation of rat depression model usingchronic unpredictable mild stress, rats within21days to accept a variety of differentunpredictable stimulated, including electric shock foot,(30times), swimming in icewater(4℃,5min),shaking (15min), thermally stimulated(5min), the tail clamp(1min), water deprivation (24h),fasting (24h)and reversed circadian. Randomlyarranged a day, average every kind of stimulation is used two times, so that variousstimuli become unpredictable long-term stress stimulation.2, Feeding on medicine in different groups: normal control group and modelgroup every day to give0.9%sodium chloride solution3ml gavage, every day bywestern medicine Venlafaxine group12.5mg/kg gavage, antidepressants GanmaiDazao Decoction scattered large, medium and small does group, respectively, everyday by38mg/kg19mg/kg9.5mg/kg orally administered.Administeredcontinuously for21days, once a day.3, Detect indicatrix: Each group before modeling, modeling, and groupadministration after treatment, respectively, they were carry by behavioral test,include open-filed test, sugar consumption test, and food consumption test, instatistical analysis. Behavioral test after completion, rapidly decapitation death of rat brain tissue samples, homogenate after high-speed centrifugation and supernatanttaken. Used with ELISA detection of neither brain monoamine neurotransmitters norepinephrine and5-hydroxyptanmine changes in the content, statistical analysis andcomparison.Results:1, Compared with the normal control group, model rats weight increase wassignificantly reduced (P<0.05); food consumption was significantly reduced(P<0.05); behavioral activity was significantly reduced (P<0.05); sugar consumptionwas significantly reduced(P<0.05); brain monoamine neurotransmitters norepinephrine and5-hydroxyptanmine content decreased obviously (P<0.05), thedifference was statistically significant.2, Compared with model group, Ganmai Dazao Decoction medium dose groupthe weight of rats increased significantly (P<0.05); food consumption wassignificantly increased (P<0.05); behavioral activity increased significantly (P<0.05);sugar consumption increased significantly (P<0.05); brain monoamineneurotransmitters nor epinephrine and5-hydroxyptanmine content was significantlyincreased (P<0.05), the difference was statistically significant.Conclusion:1, After the success model of depression, model rat brain monoamineneurotransmitters nor epinephrine and5-hydroxyptanmine content decreased,anhedonia, reduced activity, reduced ability to explore, simulate the clinicaldepression patients’ depressive state.2, Ganmai Dazao Decoction can obviously improve chronic unpredictablemild stress depression model rat behavior characteristics; significantly improveneither depression model rat brain monoamine neurotransmitters nor epinephrine and5-hydroxyptanmine levels, suggesting that Ganmai Dazao Decoction improvedispersion through the rat brain monoamine neurons transmitter5-Hydroxyptanmineand Norepinephrine levels or activity, so as to achieve the purpose of the treatmentof depression.3, Ganmai Dazao Decoction medium dose can obviously improve depression model rat behavior characteristics and significantly improve neither depressionmodel rat brain monoamine neurotransmitters nor epinephrine and5-hydroxyptanmine levels, guiding clinical rational use of drug does provide certainreference.
