Objective :To observe the effect of Lily Rehmanniae Decoction on the changes of serum anti-inflammatory cytokines and neurotransmitter in rats with chronic unpredictable stress(CUMS)depression and to explore its antidepressant mechanism.Methods :50 SD rats were randomly divided into 10 normal groups and 40 model-preparation groups.The normal group was fed normally and no stress treatment was performed.Forty rats in the other model preparation group were subjected to a 21-day depression model by chronic stress combined with solitary culture.40 rats were randomly divided into 4 groups after the model was established.,namely,normal group,model group,low dose group of Lily Dihuang Decoction,high dose group of Lily Dihuang Decoction,fluoxetine group.After the model was successfully established,except for the normal group and model group,gavage drugs were intervened for 2 weeks.The normal group and the model group were given the same amount of distilled water.On the 1st,22 rd,and 36 th days of the experiment,behavioral tests such as open-box experiments and forced swimming tests were conducted.After the last dose,the level of anti-inflammatory cytokine IL-1? in serum of each group was detected by ELISA,and immunohistochemistry was performed.The method was used to detect the expression of neurotransmitter 5-HT in the hippocampus of rats.Results:1.Behavioral index test results:Compared with the normal group,the rats in the model group had a significantly reduced number of grid passages and a prolonged immobility time for forced swimming.The difference was statistically significant(P<0.01).Compared with the model group,Lily Di Huang Tang was higher in the high dose group,the number of spontaneous activities increased significantly,and the immobility time of forced swimming decreased significantly(P<0.01).The differences were statistically significant.2.Molecular bioassay results: Compared with the normal group,the serum IL-1? content in the model group was significantly increased,and the expression of hippocampal neurotransmitter 5-HT was significantly reduced(P<0.01);compared with the model group,Serum IL-1? levels of the high dose Lily Di Huang Tang were significantly decreased in the dose group,and 5-HT expression in hippocampal neurotransmitters was increased(P<0.05).The difference was statistically significant.Conclusion:Lily Dihuang Decoction has significantly improved the behavior of chronic unpredictable mild stress depression model rats,effectively inhibit the increase of serum IL-1? levels in the rat model of chronic stress depression and increase the expression of monoamine neurotransmitter 5-HT in the brain of depression rats,suggesting that Lily Dihuang Decoction can effectively inhibit the chronic stress depression model.Increasing serum IL-1 ? levels in rats and increasing the expression of 5-HT in the hippocampus may be one of the antidepressant mechanisms.
