Clematidis Radix et Rhizoma as a kind of traditional Chinese medicine commonly used in clinical medicine, has effects of Qufeng desiccant,warming meridians and relieve pain. Clematidis Radix et Rhizoma collected in Chinese Pharmacopoeia2010comes form dry roots and rhizomes of Clematis chinensis Osbeck, Clematis hexapetala Pall, or Clematis manshurica Rupr.Besides those, other plants’roots and rhizomes are used as clematis circulating in the market nowadays. Medicinal species of clematis vary greatly from time and place and Yunan Province always uses Inula nerbosa Wall as clematis roots.Researches on the Clematidis Radix et Rhizoma mainly focus on its chemical composition and pharmacological effects, and study shows that clematis roots’chemical composition can be divided into volatile components and non-volatile components including triterpenoid saponins, flavonoids and polyphenols, alkaloids, organic acids and other non-volatile components, etc. With the research development, clematis is reported has much pharmacological activities as analgesic, anti-inflammatory, asmolysis, ntibacterial, antitumor, fall blood pressure,etc.In recent years, esearches on clematis roots’ chemical components and pharmacological effects are developing while less studies on its quality control methods. Chinese Pharmacopoeia2005only contains TLC identification but no quantitative control. Chinese Pharmacopoeia2010adds quality control by using hederagenin and oleanolic acid as indicators.After Chinese Pharmacopoeia2010’s publication, since many manufactures express that clematis in the market can not satisfy the pharmacopoeia’s standard requirement, investigation on the quality of different varieties of the clematis has great significance. Details as below:In the first chapter, the method in Chinese Pharmacopoeia2010Edition was investigated and optimized to establish the sample pretreatment methods and decide the best Chromatographic condition. The sample was pretreated by the method in Chinese Pharmacopoeia2010Edition. The optimum Chromatographic conditions are obtained as follows:the column was elected Agela Cis,the gradient elution used (A)acetonitrile-(B)water(0.1%H3PO4),the detection wavelength was205nm,the column temperature was30℃,the flow rate was1.0mL·min-1Then we determined the content of hederagenin and oleanolic acid from different species of Clematidis Radix et Rhizoma.The result shows that The content of hederagenin in most of Clematidis Radix et Rhizoma was under the limit of quantitative. Only a few Clematis chinensis Osbeck meet the quality standards for Clematidis Radix et Rhizoma in Chinese Pharmacopoeia2010Edition.In addition, we determined the content of hederagenin and oleanolic acid from both radix and rhizome in Clematis chinensis Osbeck bought in Fu Jian Province and developed the stability of the index ingredient in Clematis manshurica Rup. bought in Yongji.The second chapter established the fingerprint of Clematis manshurica Rup. R.First.the chromatographic conditions of HPLC was optimized:the column was selected Agela C18,the gradient elution used (A)acetonitrile-(B)water(0.05%H3PO4),the detection wavelength was205nm,the column temperature was30℃,the flow rate was0.8mL·min-1.Second, the method had been evaluated. The method logical evaluation showed that this method had a good repeatability, precision and stability.The fingerprints of31batches Clematis manshurica Rup. R from different areas were established.The HPLC fingerprints of samples were put into traditionanl Chinese medicine computer-aided similarity calculation software,control fingerprint was fit out,and then the similarity of each sample compared with the control fingerprint was calculated.Then we established the fingersprints of10Clematis chinensis Osbeck in the same method.At last,seven triterpenoid saponins molecular weight of main marks were obtaind by LC-MS.In our research, Quality survey of different species of Clematidis Radix et Rhizoma was made by determining the content of hederagenin and oleanolic acid from Clematidis Radix et Rhizoma. Besides,We esteblished the fingerprints of31batches Clematis manshurica Rup. from different areas.it is significant to standardize Chinese traditional medicine market.
