Herba Saussureae Involucratae is our precious medicinal traditional Chinese medicine (TCM), which has good effects on dispelling cold, strengthening yang, enriching blood and warming the uterus. Xuelian for Injection is pure TCM simple preparations of Herba Saussureae Involucratae, which has the advantages of definite therapeutic effect and security as Xuelian Injection, however, it shows a high stability. From the view of overall control of Herba Saussureae Involucratae and Xuelian for Injection, this paper preliminarily deduced chemical substances base of water soluble components in Herba Saussureae Involucratae, then studied the fingerprints of Herba Saussureae Involucratae and Xuelian for Injection.1. HPLC-HR-MS/MS was employed to qualitatively analyze the chemical substances of water soluble components in Herba Saussureae Involucratae. Twenty chemical substances were assigned or deduced, including chlorogenic acid and its isomers and derivatives, nucleosides, phenylpropanoid, amino acid, which established the foundation of studying physiological process, clarifying pharmacodynamic substances and action mechanism.2. HPLC fingerprints of whole grass and flower, bract, leaf, stem were established. The software of "Digitized Evaluation System for Super-information Characteristics of the Traditional Chinese Medicine Fingerprints 3.0”was adopted for the system evaluation of the fingerprints. Using the referential fingerprint as a standard, systematic quantified fingerprint method described the quality of ten batches. Two batches were better, two batches were good, and others were fine. By comparing the fingerprints of whole grass and different parts, it was proved that there was a large discrepancy between them in terms of the kinds and contents of their chemical substances, so a certain part could not substitute the whole grass for using as medicines.3. Fingerprints of Xuelian for Injection were established under the wavelength of 280 nm by HPLC, which were all best evaluated by systematic quantified fingerprint method. Using macro qualitative similarities Sm and macro quantitative similarities Pm as indexes, the differences between the fingerprints of before and after various damages, such as light, high temperature, acid, base and oxidation, were studied, which provided new references for overall control of Xuelian for Injection under the conditions of production, transportation and storage.4. The triangle and tetrahedron optimization methods were developed for the selection of back ground electrolyte (BGE) in capillary zone electrophoresis (CZE). Chromatographic fingerprints index F was used as objective function to judge the separation and the best BGE was selected. HPCE fingerprints of whole grass and flower, bract, leaf, stem were established, which were evaluated by systematic quantified fingerprint method. The contents of chlorogenic acid and rutin were determined simultaneously by internal standard method.
