Object:this article discussed the differential expression rule of rat’s ventricular muscle proteome after4weeks aerobic exercise and primarily screened out some proteins related to exercise adaption for further research in the effect of aerobic exercise on myocardial proteome and the improvement of heart function.Method:20SD rats were divided into control group (CG) and experimental group (EG) according body mass and exercise capacity. The EG performed incremental treadmill training with the speed up to24m·min-1and40min duration. The EG was executed after anesthesia6hours later after exercise with CG at the same time. The ventricular muscle’s whole protein of two groups was abstracted for two dimensional gel electrophoresis separation and the2-DE gels were analyzed with Image Master2D Platinum. After analysis, the new emerged and that disappeared spots and the differentially expressed amount more than5folds spots were selected for identification with time-of-flight tandem mass spectrometry and quantitative analysis with RT-PCR.Result(1) An average of478±7protein spots were detected in EG and563±24in CG and45protein spot were differentially expressed and distributed in WM of10~70kD and pH of5~8;(2) The number of up-regulated protein spots more than2folds is27and that of down-regulated to1/2is16. there are’also7spots up-regulated more than5folds and5down-regulated to1/5. The mass spectrometry identified12protein spots from selected protein spots. They are alpha actin, pyruvate dehydrogenase E1alpha1, phosphorylated triose isomerase, mitochondrial election transfer flavoprotein alpha subunit, Translationally-controlled tumor protein, Ubiquitin-conjugating enzyme E2N,4-three methyl tryptophan amino aldehyde dehydrogenase, mitochondrial succinate dehydrogenase coenzyme flavoprotein subunit, serum protein, selenium protein1, mitochondria branched chain amino transferase, ubiquitin fusion degradation1-like. RT-PCR results show that the mRNA amount of alpha actin,, selenium binding protein1, translationally controlled tumor protein, ubiquitin conjugating enzyme class E2, mitochondrial electron transfer flavoprotein subunit A,4-three methyl tryptophan amino aldehyde dehydrogenase, phosphorylated tiiose isomerase on EG are exceed CG (P<0.05) and that of mitochondria branched chain amino transferase is lower (P<0.05).Conclusion:After4weeks aerobic exercise, rat’s ventricular muscle proteome was obviously changed. Myocardial energy metabolism raised and the EG’s left ventricular muscle produced adaptive change. This research has screened out8proteins which have not researched in the filed of sports medicine. The RT-PCR result showed that the expression in protein level and gene level of8proteins is same except selenium protein1.
