The purpose of this study is to discuss the influence which auricular muscle proteomics on increasing progressively exercise-load-training,sieve targeted protein of variability expressed and important significance in sport, investigation rule of development and effluence of sports fatigue in proteome.Applying theory and technique of proteome,ten male SD rats were divided into two groups randomly :the rest group (5) and the exercise group (5). After increasing progressively exercise-load-training 7 weeks,exhausting training 6 hours,they were narcotization executed at the same time. Extracted whole protein of auricular muscle,the proteins were separated by 2-DE,stained by Coomassie brilliant blue dye,analyzed by MALDI-TOF-TOF-MS in turn.354±40 and 382±24 protein spots were found in the sport group and test group, matching rate were 68.67%±9.87% and 72.67%±8.62%,coefficient correlation were 0.71;in the 104 differentially expressed spots after traning,66 in up-regulation 2 times,including 18 in up-regulation 5 times, none protein spot in increase; 38 in down-regulation 2 times,including 23 in down-regulation 5 times,7 in obsolescented. Selected protein spots of differential expressed more 5 times as object proteinum, there wre mass chromatographic analysis, 5 protein spots were assessmented: myosin light polypeptide 4 and aspartate aminotransferase were up-regulation, Myosin-6 was down-regulation and absence.Experimental result indicated that auricular muscle protein were obviously variability expressed, and it was sufficient experiment base in order to profoundly study original objective proteinum of sports fatigue. The auricular muscle protein of variability expressed are related to signal transduction, energy metabolism, substance metabolism and myocardial function. The disappeared and dowrn-regulated proteins in exercise group are ralated to decrease of muscle contraction and motor ability and proceed of sports fatigue;the up-regulation are ralated to injury of histiooytic in long-term increasing progressively exercise-load-training. Most important, myosin light polypeptide 4 and aspartate aminotransferase that successfully sieve through method of proteome will impact organism disadvantaged, they are not investigation yet ,and it will be offer new thought in study domain of sport heart.
