Objective:The objective of this study is to investigate the effects of training on IGF1-IGF1R of spontaneously hypertensive rats(SHR), and to explore the potential mechanism and preventive effect of hypertension and its complications under the condition of appropriate exercise provide the theoretical basis of prophylaxis and treatment of hypertension and its complications.Methods:The sixteen male Wistar rats were randomly divided into the two groups of eight each:Wistar control group (WC)and Wistar training group(WT); The sixteen male spontaneously hypertensive rats were randomly divided into the SHR control group (SC)and SHR training group(ST); WT and ST groups carried out90minutes swimming6times every week during the experiment.Determined the rats blood pressure and the body weight each week during experimental period,after eight weeks determined Insulin, glucose、AngⅡ and IGF1in blood, AngⅡ、IGF1、IGF1R and MAPK of the VSMC in Aorta.Results:1. After daily90minutes swimming for8weeks, we found the weight of SHR training group obviously descended more than that in control Group, It means that exercise can inhibit the increase of weight in SHR rats.The blood pressure in SHR control Group obviously raised more than before experiment (P<0.01), the SHR control group, elevated blood pressure range of significantly reduced (P<0.01) indicating that regular exercise can delay SHR rat blood heighten.2. After daily90minutes swimming for8weeks, we found the Ang Ⅱ in aortic and blood Of SHR control group are more than Wistar control group, explain hypertensive rats blood and aortic of Ang Ⅱ higher than normal rats.the Ang Ⅱ in plasma and aortic of SHR training group significantly reduce than SHR control group (P<0.05), indicating that regular exercise can significantly reduce the Ang Ⅱ content in plasma and the aortic by SHR rats.3. After daily90minutes swimming for8weeks,we found the insulin resistance index of SHR control group significantly higher than Wistar control group (P<0.01), indicating that hypertension has obviously insulin resistance. While the insulin resistance index of SHR training group significantly reduced than SHR control group (P<0.01), indicating that regular exercise can improve insulin resistance status in rats. 4. After daily90minutes swimming for8weeks,we found the IGF1in aortic and blood Of SHR control group are more than Wistar control group (P<0.05), explain hypertensive rats blood and aortic of IGF1significantly higher than normal rats. While the IGF1in blood and aortic of SHR training group are more than SHR control group (P<0.01), indicating that regular exercise can reduce the IGF1in blood and aortic.5. After daily90minutes swimming for8weeks, we found the SHR control group are significantly higher than Wistar control group of IGF1R in aortic (P<0.05), it is means IGF1R in aorta is significantly increased in hypertensive rats. While the IGF1R in aortc of SHR training significantly reduced than SHR control group (P<0.01), indicating that regular exercise can reduce the aorta IGF1R content of SHR rats.6. After daily90minutes swimming for8weeks,we found the MAPK in aortic of SHR control group are more than Wistar control group (P<0.05),it is mean how high blood pressure can cause MAPK content increased.While the MAPK in aortic of SHR training group significantly reduced than SHR control group(P<0.05),That regular exercise can reduce the MAPK in aortic of SHR.Conclusion:1.90minutes swimming for8weeks can significantly inhibited suitable SHR rat blood of hypertension further raise, have certain soothing effects, meanwhile, exercise on Wistar rats and the growth of SHR rat weight has certain inhibition;2.90minutes swimming for8weeks can effectively reduce for IGF1content and IGF1R content, inhibiting proliferation of vascular smooth muscle, has a positive effect to slowing hypertension course and reduce high blood pressure;3. IGF1-IGF1R in hypertension plays an important role, sports can effectively reduce IGF1content and IGF1R content, lower blood pressure, delay hypertension course. Its mechanism is:90minutes swimming for8weeks reduce hypertension in rats Ang II content and improve insulin resistance, reduce the release of IGF1state, restrain the vascular smooth muscle in the content of IGF1R expression, MAPK activity reduced, inhibit vascular smooth muscle proliferation, reducing blood pressure.
