Experimental Objective:In order to explore intervention effects of aerobic exercise or/and soy peptides (SPI) on the informing of high-fat-diet-induced insulin resistance IR) in SD rats and its mechanisms.Empirical method:The test rats were randomly divided into 5 groups: Ordinary feed control group (C group), high fat diet group (HF group), high fat diet + exercise group (HE group), high fat diet + soy peptides group (HS group), high fat diet + exercise + soy peptides group (HSE group). The food quantity in each group were determined every day, and body length andthe body weight were surveyed each week. While the SD rats were fed high fat diet to form IR, aerobic exercise training was conducted or / and SPH was supplememted. The exercise plan was 60rain's swimming without a heavy load for 10 weeks, 6 days per week. The dosage of SPI filled the rats stomach was 500mg/kgBW. The oral glucose tolerance (OGTT) were carried on at the 8th test week. Fasting plasma glucose (FPG), fasting plasma insulin (FINS), triglyceride (TG), total cholesterol (TC), the blood serum tumor necrosis factor-alpha (TNF-α) and interleukin-6(IL-6), C peptide(CP) were determined at the end of the experiment, and insulin sensitivity index (ISI), homeostasis model assessment-IR index (HOMA-IR), Lees index (Lees) were calculated to judge the formation of IR as well as the intervention effects of aerobic exercise and SPI on IR.Experimental results:(1)Compared with C group, FINS, FPG, HOMA-IR in the HF group increased significantly (P<0.05, P<0.01), but ISI reduced obviously (P<0.01); The OGTT indicated blood glucose level in HF group were obviously higher than those in C group before meat and after the 2 hours meal(P<0.01, P<0.05). Simultaneously serum TG, TC and IL-6 contents in HF group increased remarkably (P<0.05, P<0.01), although serum TNF-αcontent in HF group rats was non-significance difference with that in C group, its ascension scope reached 25%. Compared with group C, the serum CP content in HF group was non-significance difference.(2)Through the double factor variance analysis, aerobic exercise or supplemented SPI had remarkable intervention on the formation of IR induced high fat diet in rats. While the high fat diet induced the rats IR formation, aerobic exercise training was carried on which might cause FPG, FINS, HOMA-IR in the high fat diet rats to reduce obviously, but ISI increased remarkably (P<0.05, P<0.01). At the same time, it caused serum TG, TC, TNF-α, and IL-6 concentrations as well as blood glucose before meal and after 2 hours meal in high fat diet rats to reduce significantly(P<0.05, P<0.01). While the high fat diet induced the formation of IR in the rats, SPI supplement might cause FINS, HOMA-IR in the high fat diet rats to reduce obviously, and ISI to increase remarkably (P<0.01). It also caused serum TG, TC, TNF-αIL-6 contents and blood glucose before meal, after the 2 hours meal in the high fat diet rats to reduce obviously (P<0.05, P<0.01). The FPG content reduced, but there wasn't significant difference. Aerobic exercise or supplemented SPI might cause serum CP content in the high fat diet rats to reduce, but there wasn't significant difference.(3)Through the double factor variance analysis, while aerobic exercise united SPI supplement might cause FPG, FINS, HOMA-IR in the high fat diet rats to reduce, ISI to increase, but there wasn't significant difference. In the OGTT, it caused blood glucose before meal and after the 2 hours meal in the high fat diet rats to reduce obviously (P<0.01). At the same time, the exercise and supplemented SPI might cause serum TG, TC, TNF-α, IL-6 contents in high fat diet rats to reduce obviously, and it caused serum CP content to increase remarkably (p<0.01).Experimental conclusion:(1)10 week-long high fat diets might cause FPG, FINS, HOMA-IR to increase remarkable, but ISI reduced obviously, it indicated that 10 week-long high fat diets might induce the formation of IR in rats.(2)While the high fat diet induced rats forming IR, 60min' swimming or supplemented SPI was carried on for 10 weeks that might reduce FPG, FINS, the HOMA-IR and enhance the ISI in the high fat diet rats obviously. It indicated that 60min' swimming or supplemented SPI for 10 weeks might prevent the formation of IR induced high fat diet obviously. Moreover, aerobic exercise united supplement SPI had the promoter action to prevent the formation of IR in the high fat diet rats to a certain extent.(3) The mechanisms of IR formation induced high fat diet in rats were related with increase of blood lipids, decrease of the circumference Ins R sensitivity, as well as increase of TNF-α, IL-6 secretion in high fat diet rats. But aerobic exercise or supplemented SPI might prevent the formation of IR induced high fat diet through the improvement of blood lipids metabolism, the enhancement of the sensitivity of Ins R in circumference organize as well as the suppression of TNF-α, the IL-6 secretion.
