| Objective:Based on the theory of Chronobiology, establishing the model of type2diabeticrats,and put into aerobic Chronoexercising according to blood glucose fluctuation inrhythm characteristic,to explore the changes of SAA Tanis system in type2diabetesmellitus rats and the effects of aerobic Chronoexercising to the IL-6,SAA and it’sreceptor Tanis protein.Methods:1.80healthy and clean SD male rats,all of them were fed with the regulardiet one week to adapt to the environment.One week later,divided them into twogroups randomly:normal control group(Group N,12),model establishing group(Group F,68).Group F was fed with the high fat high glucose diet for two weeks,Group N was fed with the regular diet for four weeks.At the end of the fourth week,after fasting12hours,the weight of the rats were measured.According to30mg/kg body weight,the F Group rats were injected with STZ on stomach for twice.The similar dosage of citrate buffer solution was given to N Group rats.24hourslater,we tested the blood glucose of F Group rats,if the result of the blood glucose was higher than or equal with16.7mmol/L,it was succeed to establishing thetype2diabetes mellitus rat model.After that,F Group rats were given high fathigh glucose diet for one week,then we tested the blood glucose of the F Grouprats by tail vein again56rats modeling success, the successful rate was83%.2.The56rats were divided into two groups randomly:diabetes control group(Group C,14),exercise group(Group S,42).Then the S Group were divided into three groups randomly:exercise at six am(Group SM,14),exercise at twelve(Group SN,14),exercise at six pm(Group SE,14).S Group did adaptability treadmill running 1week,then put into aerobic chronoexercising according to the plan.On the sixth weekend,all rats fasted for12hours and then measured the blood glucose by the tail vein,separated the plasma of apex blood for further test.Take off the muscle and the right of the liver,then put it into liquid nitrogen rapidly.Last,put themin the fridge.The temperature of the fridge is Minus seventy degrees Celsius forthe further test;Take off the pancreas,then being fixed in10%formalin solutionfor the further test.3.we tested the FINS,IL-6,SAA,Tanis protein by ELISA.Observe the pancreatic pathology by microscope.4.All the data were analyzed using the SPSS17.0software.Result:1.The blood glucose of N group rats at18:00significantly higher than6:00(P<0.01),plasma insulin have no significant difference between18:00and6:00(P>0.05).2.The FPG of T2DM is above16.7mmol/L,and have no significant difference between18:00and6:00(P>0.05);FINS,HOMA IR increased(P<0.01);IAI,Homa βdecreased(P<0.01);IL-6,SAA,increased(P<0.01);The level of the Tanis protein increased(P<0.01).3.After six weeks of aerobic Chronoexercising,FPG level of S group(SM group,the SN group and SE group) were lower than C group(P<0.05);The SN,SE group was lower than SM group(P<0.05).Compared to the C group,FINS of S group(SM group,the SN group and SEgroup) were lower than C group(P<0.05),but no significant difference between the SM,SN,and SE group(P>0.05).After six weeks of aerobic Chronoexercising,compare to the C group,HOMA-IR in group S group decreased(P<0.01),HOMA β of S group increased(P<0.01),but it didn’t influent the level of IAI(P<0.05).4.After six weeks of aerobic Chronoexercising,compared to the C group,the level of IL-6and SAA of S group(SM group,SN group,and SE group) were lower than C group(P<0.01),but the level is higher than N group(P<0.01);The SE,SN group was lower than SM group significantly(P<0.01).5.After six weeks aerobic Chronoexercising,the level of the Tanis protein inthe liver and muscle of the S groups(SM,SN,SE group) were lower than C group significantly(P<0.01);The level of Tanis protein in the liver and muscle of theS groups were lower than N group(P<0.05);The level of Tanis protein were no significant difference between the SM,SN,and SE group(P>0.05).Conclusions:1.The blood glucose level fluctuations of the healthy rats have a significantcircadian rhythm,the structure of the pancreatic tissue is complete. The T2DM rats fluctuation in blood glucose levels is dysrhythmia,exsiting insulin resistance andlow grade inflammation,the pancreatic tissue structure partially damaged.2.The six weeks of Aerobic Chronoexercising can effectively reduce FPG,improve insulin resistance.The effect of aerobic Chronoexercising in FPG peak hours (18:00) or12:00pm were better than in valley hours(6:00), but no effect on the biological rhythm of the fluctuations in blood glucose.3.The six weeks of Aerobic Chronoexercising can downregulate the level ofthe plasm inflammation factor and SAA Tanis system,ease the body’s inflammatoryresponse,improve insulin resistance of the liver and protect it from damage.Theeffect of aerobic Chronoexercising in FPG peak hours(18:00) or12:00am werebetter than in valley hours(6:00). |
