Objective:Based on the theory of Chronobiology, build the model of type2diabetic rats, andput into aerobic Chronoexercising according to blood glucose fluctuations in rhythmcharacteristics, to explore the Changes of cruor-fibrinolysis system in T2DM rats andthe effects of aerobic Chronoexercising to the specificity index of cruor-fibrinolysissystem: P-selectin, TAT, t-PA and PAI-1.Methods:1. Got80healthy, clean,4-week-old,180~200g weight, male SD rats, adaptabilityfed about1weeks。Then randomly divided into the health control group (N=12, H) anddiabetic model group (N=68, G). H group fed routine feedstuff, the G group was givenhigh sugar high fat diet. On the4-th weekend, Group G rats were fasted for12hours,and then were injected of STZ (30mg/kg) under the lower left cavum abdominis onceevery other day for two consecutive times; H group were injected with an equivalentdose of placebo. Measure the fasting plasma glucose(FPG) of the tail vein after24hoursof the2ndinjection STZ. After the model was successful (FPG≥16.7mmol/L),continued to fed for high glucose and fat one week. On the5-th weekend measured theFPG of the tail vein again.56rats modeling success, the successful rate was83%.2.56T2DM rats were randomly divided into diabetic control group (N=14, C)and exercise group (N=42, S), and the exercise group were divided into6:00group (N=14, the SM group),12:00group (N=14, SN) and18:00(N=14, SE). S group didadaptability treadmill running1week, then put into aerobic exercise chronoexercisingaccording to the plan. On the6-th weekend, each group fasted for12hours and then measured the FPG of the tail vein, separated the plasma of apex blood for further test.3. ELISA method for measuring FINS, TAT, P-selectin, t-PA and PAI-1content.4. Process the test data by SPSS11.5.Result:1. The blood glucose of health rats at18:00significantly higher than6:00(P<0.01),plasma insulin have no significant difference between18:00and6:00(P﹥0.05).2. FPG of T2DM rats≥16.7mmol/L,and have no significant difference between18:00and6:00(P﹥0.05); FINS, P-selectin and TAT and PAI-1increased (P <0.01), t-PAdecreased (P <0.01); Homa-IR increased(P <0.01), IAI Homa-β decreased (P <0.01).3. After six weeks of aerobic Chronoexercising, FPG level of S group (SM group,the SN group and SE group) were lower than C group (P <0.05); the SN, SE group waslower than SM group (P <0.05).Compared to the C groups, FINS of S (SM group, the SN group and SE group)were lower (P <0.05), but no significant difference between the SM, SN, and SE group(P>0.05).4. After six weeks of aerobic Chronoexercising, compare to the C group, Homa-IRin group S group decreased (P <0.01), Homa-β of S group increased (P <0.01),but theIAI have no change.Also plasma P-selectin, TAT and PAI-1levels in S group (SM group, the SN groupand SE group) were decreased (P <0.01), plasma t-PA increased (P <0.01). SE groupchange most significantly (P <0.01) in SM, SN and SE groups.Conclusions:1. In normal state, the level of FPG express circadian rhythm. The FPG hadincreased in T2DM rats but circadian rhythm was disorder, accompanied with insulinresistance;function of cruor-fibrinolysis system imbalance and blood was inhypercoagulable, prothrombotic state.2. The Aerobic Chronoexercising can effectively reduce FPG, control weight andimprove insulin sensitivily. The effect of aerobic Chronoexercising in FPG peak hours(18:00) or12:00pm were better than in valley hours (6:00), but no effect on the biological rhythm of the fluctuations in blood glucose.3. The Aerobic Chronoexercising can improve blood hypercoagulability,prothrombotic state in T2DM rats, and tend the balance between the function of plateletand the function of cruor-fibrinolysis system, to ensure blood flow smooth.
