This paper has studied the best culture conditions of laccase production about Myrothecium verrucaria GH-01with the shaking culture and the enzyme’ production of GH-01in fermentater. And we also has studied the decolorization about GH-01to4kinds of dye (anthraquinone dyes:alizarin red, fang methane dye:fuchsine, azo dyes:Congo red, other types of dyes, methylene blue) in co-culture condition. Then we has obtained some results in the paper are as followed:(1) In the shaking culture conditions, there is1.2times of fermentation medium in our medium which has potato200g,waterl L,glucose24g, peptone48g, monopotassium0.36g, and0.18g magnesium sulfate in28℃with the2mM Cu2+. In this condition, the peak value of GH-01with shakeing culture could be more than100000U/L which may be repeatable. With15L fermentater, the2nd peak of GH-01is higher than the1st one. It is failed to reach the peak of enzyme activity.(2) We has got GH-01pure laccase with ammonium sulfate chromatography,dialysis, SDS-PAGE, DEAE Sepharose Fast Flow anion exchange chromatography, SephadexG-75chromatography, Native-PAGE. We also researched the decolorization ability of4typical kinds of categories..The results show that the decolorization of ACE and VA of Congo has promoted a evident role with the increased to80%;. Fuchsine and alizarin red almost not influenced by media, decoloring rate basic remain unchanged; Our HBT with methylene blue decolorizing effect, decoloring rate from original54.67%to63%.(3) Under the optimum culture conditions of GH-01,we has studied the decolorization of different concentrations of the above four dyes in the co-culture conditions. Decolorization of the final concentration of10mg/L methylene blue is the best, decolorization rate increase to87%; The final concentration of75mg/L of alizarin red decolorization effect also good, decolorization rate reach80%; different final concentration of Congo red and fuchsine decolorization effect are not ideal, the decolorization rate is only50%.
