The genus Pestalotiopsis is a major group of endophytic fungi in plants. They were significant resources for producing a wide range of chemically diverse metabolites which were anticancer and antibiosis. The endophytic Pestalotiopsis on plants belonging to Fagaceae, Lauraceae and Palmae were investigated, and the diversity was carried out. The phylogenetic trees were generated based on ITS and EF1-a sequences, which were recognized there were relationship between phylogenetic evolution and morphological characters of Pestalotiopsis. The main results were summarized as follows:1-, The diversity of endophytic Pestalotiopsis on the tree plantsA survey of endophytic Pestalotiopsis on22plant species belonging to Fagaceae, Lauraceae and Palmae was carried out. A total of161Pestalotiopsis isolates belonging to30endophytic Pestalotiopsis species were isolated, of which17、19and17endophytic Pestalotiopsis species, and Pestalotiopsis microspora was dominant species in this investigation. Eight、twelve and twelve endophytic Pestalotiopsis species were respectively obtained from Cyclobalanopsis fleuryi (Fagaceae), Cinnamomum camphora (Lauraceae), Arenga engleri (Palmae) representing plant of every families which the most endophytic Pestalotiopsis species hosted on. The colonization frequencies of endophytic Pestalotiopsis on the tissues of twigs and leaves were3.07%and0.41%, respectively and the diversity analysis indicated that endophytic Pestalotiopsis was more plentiful on twigs than leaves of these plants. The colonization frequencies of endophytic Pestalotiopsis in spring, summer, autumn and winter respectively were3.44%、1.53%、1.1%and1.75%, and Shannon-Wiener indexes of endophytic Pestalotiopsis in spring, summer, autumn and winter respectively were2.5006,2.2902,2.2475and1.9541, and it indicated that the colonization frequencies and species diversity of endophytic Pestalotiopsis were the highest in spring compared with other seasons.2、Pestalotiopsis genus of molecular phylogenetic relationshipThe molecular phylogenetic relationship of Pestalotiopsis based on EF1-a sequences was firstly studied and the result showed that phylogenetic trees generated from ITS sequences and EF1-a sequences were very similar. The species of Pestalotiopsis were partitioned into2groups pale (intermediate cells were concolorous, olivaceous to pale brown) and dark (intermediate cells were versicolorous, umber to fuliginous) on phylogenetic trees based on ITS and EF1-a sequence and it indicated that the pigmentation of median conidial cells of Pestalotiopsis were the primary character on taxonomy. A clade was divided into seven subclade(A1、A2、A3、A4、A5、A6、A7) and B clade was divided into two subclade. The morphological charaters of condia were unique in every subclade of A clade on apical appendage knobbed at the extremities, or number of apical appendage, or apical appendage single then branched, or position of the apical appendage arising from apical hyaline cell. The unique morphological charater of condia which distinguished B1and B2was apical appendage knobbed at the extremities or not.It showed the character of apical appendage knobbed at the extremities or not was an important character in dividing the dark group. The phylogenetic tree of EF1-a was divided into the clades of A3、A4、 A6、A7becaused of deficiency of representing strains of Al、A2and A5on ITS tree. This result indicated that ITS and EF1-α sequences were promising on being chosen as molecular barcode.
