The black resin of Aquilaria sinensis is called Chinese agarwood which is a famous traditional medicinal material,and is an important material for traditional spices or crafts.'Qi-Nan' is the most expensive and precious kind of agarwood,'Qi-Nan' has more resinous content than other agarwood of lower quality,its fragrant odor that could be obviously smelt?Phytochemical studies on the the Chinese agarwood and the endophytic fungus of Chinese agarwood have previously led to the isolation of a series of compounds,and some of which possessed biological activities.In an effort to search for bioactive compounds,on the basis of previous work,chemical constituents and bioactivities from the endophytic fungi of high quality agarwood(named 'Qi-Nan' in Chinese)were studied.The compounds were isolated and purified by various column chromatographic techniques,and their structures were identified through a combined analysis of physicochemical properties and spectroscopic evidence(VU,IR,MS,1D and 2D NMR).The antimicrobial and cytotoxicity activities in vitro of the compounds were evaluated by paper disc diffusion method and MTT method,respectively.Moreover,the compounds were also tested for inhibitory activity against acetylcholinesterase using the Ellman colorinetric methodTwenty-nine compounds were isolated from the endophytic fungus Fusarium sp.HP-2 of 'Qi-Nan' eaglewood,and their structures were determined as:5,9-dihydroxy-2,4-dimethyl-8,9-dihydronaphtho[I,2-b]furan-6(7H)-one(1),9-hydroxy-6,8-dimethoxy-10-(2-oxopropyl)-3,4-dihydro-1,4-methanobenzo[c]oxepin-5(1H)-one(2),dihydronaphthalenone(3),fusamaphthoquinone A(4),(+)-solaninol(5),2,3-dihydro-5-hydroxy8-methoxy-2,4-dimethylnaphtho[1,2-b]furan-6,9-dione(6),4-hydroxy-5-methoxy-3-methyl-4,5,5a,7,8,8a-hexahydro-lH-pentaleno[1,2-c]pyran-1,6(3H)-dione(7),Fusarpyrone B(8),Fusarpyrone A(9),5-hydroxy-4-hexanolid(10),10,11-dihydrocyclonerotriol(11),lumichrome(12),2'-O-methyluridine(13),N-(2-phenylethyl)acetamide(14),nicotinic acid(15),Uracil(16),monaspilosin(17),methyl-2-hydroxy-3-(4-hydroxyhenyl)propanoate(18),Methyl-2-(2-methoxyphenyl)acetate(19),methyl 2-(2-hydroxyphenyl)acetate(20),methyl 2-(4-hydroxyphenyl)acetate(21),4-(2-hydroxyethyl)phenol(22),1-(4-Hydroxyphenyl)ethane-1,2-diol(23),2-Methoxy-2-(4'-hydroxyphenyl)ethanol(24),serratenediol(25),5?,8?-epidioxy-(20S,22E,24R)-ergosta-6,22-dien-3?-ol(26),stigmasta-7,22-diene-3?,5?,6?-triol(27),10-Hydroxydodecanoic acid(28),and methyl linoleates(29)?Amog them,Compoud 1 and 2 are new compouds,compoud 7 is a new skeleton compound.Comound 5,6 and 28 showed inhibitory effect against Ralstonia solanacearum,and comound 28 revealed inhibitory effect against Staphylococcus aureus.Compound 5,6 and 26 exhibited cytotoxic activities in vitro against K-562,BEL-7402 and SGC-7901 tumor cell line.Compound 2,4,5,6,21 and 27 showed the inhibitory activities against acetylcholinesterase.
