The flower buds of Chrysanthemum indicum L are widely used in heat-clearing, detoxifying, improving eyesight, protecting liver, decreasing blood pressure and protecting the cardiovascular. Flavonoids are a main kind of components with a wide range of bioactivities in the flower buds of Chrysanthemum indicum L. In this dissertation, the leaching and extraction process of total flavonoids from wild chrysanthemum L was studied, and the separation of flavonoid monomer utilizing high speed counter current chromatography (HSCCC) was explored.The main contents are as follows:1. The leaching process was studied in order to maximize the leaching result through investigating the influences of ethanol concentration, ratio of solid to liquid, temperature, leaching time and leaching times on the yield of the total flavonoids, and the optimal leaching conditions were obtained;.2. The liquid-liquid extraction process was studied in order to maximize the extraction result. a suitable extractant was chosen and the influences of feed concentration, phase ratio and extraction times on the yield of the total flavonoids and purity were investigated, and the optimal extraction conditions were obtained. Furthermore, it is found that the purities of extracts obtained at the second and third extraction are higher than60%, thus these extracts can be used for the separation of flavonoid monomer.3. High speed counter current chromatography (analytical and preparative) was utilized to further purify the extracts. Retention of the stationary phase of solvent system n-hexane:EtOAc: MeOH:water in different revolution speed and flow rate in analytical HSCCC was investigated, and it is found that in analytical HSCCC, the proper revolution speed is above1400rpm and proper flow rate is0.5-1.0mL-min4. Analytical HSCCC was used to choose proper solvent system from various ones with different volume ratio, and the chosen solvent system was applied to preparative HSCCC in scale-up separation test.The result showed that solvent system n-hexane:EtOAc:MeOH:water can be a valid choice in separation of flavonoid monomer, especially in preparative HSCCC which has higher resolution. In the experiment, the flavonoid monomer was not obtained, and the reason was analyzed while the improving suggestion was given.5. In the basis of optimal leaching and extraction conditions, pilot plant test of200times scale-up was conducted. The design of pilot plant has taken full account of materials recycling and waste disposal, with a simple preparation process, materials recycling, less solvent consumption and less waste discharge.
