Collagen, a protein that is the highest amount in all multicellular animals, is one of the most important parts in connective tissue of human body. Because it determines the tenacity of connective tissue, it plays an important role that connects tissues in animal body. Selenium polysaccharide can increase the phagocytosis of macrophage. It has different degree of influence for the processes of chemotaxis, swallow and killing in macrophage. Combining collagen and immune globulin can significantly improve the immune function of human body. Selenium polysaccharide can stimulate the immune globulin and the production of antibodies, and improve the sterilization ability of macrophage. Combining collagen and selenium polysaccharide, can significantly improve the body’s immunity against foreign virus, and enhance the ability of resistance to disease, to improve the quality of people’s life.Conclusions could be summed as follow:1. Study on the preparation process of Se-enriched polysaccharide collagen granulesThe article take full advantage of materials to prepare the particles included the collagen extracting from the skin of codfish, selenium-enriched carrageenan, xylitol, sodium carboxymethylcellulose and water. Through the experiments, we study the best conditions of preparing Se-enriched polysaccharide collagen granules. Define DLL as the proportion of the particles that can pass20mesh sieve while can’t pass40mesh sieve in total particles after drying.(1) First, single factor experiments were made to find out how each factor affects DLL. Make sure that collagen, selenium-enriched carrageenan and water are the three main factors. (2) Then using response surface methodology based on a three-factor central composite design. Make sure the best adding conditions of three main factors to prepare the Se-enriched polysaccharide collagen granules.The best amounts of the added collagen peptide, selenium-enriched carrageenan,sodium were optimized to be23.63%、31.87%、12.87%.2. Effects of Se-enriched polysaccharide collagen granules on immune function in mice.By using the above optimum conditions of preparation of granules we study the spleen index, thymus index, cell proliferation and the activity of NK cells in mice and analysis of its influence on cell immunity of mice Comprehensively.Explore the effect of Se-enriched polysaccharide collagen granules to NK cells prolif--eration condition and activitice, Achieve probe into the effect of the regulatory role of Se-enriched polysaccharide collagen granules to NK cells immunity activity purpose. Methods:the method for the determination of the organ/ratio, MTT colorimetric method and the method for the determination of the activity of NK cells.Result:(1) The thymus and spleen coefficient of mice that is functioned by Se-enriched polysaccharide collagen granules is higher than the control group,and With the increase of dose the effect will have increased. This indicates that Se-enriched polysaccharide collagen granules can affect immune function at a certain extent.(2) The Se-enriched polysaccharide collagen granules on immune cell in mice proliferation plays a significant inhibitory effect.(3) When E/T ratio is20/1and50/1, Se-enriched polysaccharide collagen granules for the killing rate of immunocyte on mice has certain effect, especially for the low dose. And the effect at the ratio of E(Effect) to T(Target) as50:1is higher than at the ratio of E(Effect) to T(Target) as20:1.(4) By means of the test results of appreciation and kill, Se-enriched polysaccharide collagen granules has certain enhancement nonspecific immunity function of Non-specific immune. The granules made by compounding Se-enriched polysaccharide and collagen with the most suitable amount of addition will improve the organism immunity better.
