It resists cancer, inflammatory, oxidation, blood fat, virus and hance immunity etc.So it is a safe,effective drug,and has a good clinical application future.However, the domestic preparation study is limited.Troche and capsule are common, new forms are rare.In addition, curcumin is unstable towards light and heat, can be metabolize by enterohepatic circulation and bioavailability is very low orally.Therefore,we choose collagen as a carrier material, wrap curcumin with collagen through emulsification condensation method to make a new kind of pharmaceutical dosage form-microparticles,which aimed at raise the bioavailability of curcumin in body and have the best therapeutic purpose.Firstly, we have a comprehensive study on extracting, puring and appraising of the new drug carrier-collagen.Fresh rat tail tendon is took as raw material,add 0.05MTris/HCL(PH7.5) containing 1MNaCl for fore-dealing and eliminating the impurity.Then dip tendon in 0.5M HAC for 4 days,mix round 6 times each day,maintaing 4℃. Centrifuge the mixture by high speed freezing centrifuge and the supernatant is crude collagen aqueous solution. Then salt it out and centifuge to obtain the sediment, which was dialysed by distilled water. The concentration of obtained refined collagen aqueous solution is mensurated by UV.Mensurate the max absorbed wavelength of collagen aboved and the standard collagen(type I) by UV scanning.The result are the same,which is 205nm.Then use SDS-PAGE to study the above two.The result show that the characteristic bands of them are equal in main.The molecular weight of collagen are 100000,97400,43000,31623.We obtain one characteristic band of collagen by IFE ,the PI of which is 4.90.At last,determine by uranyl buffer salts,which show that the freeze-dried collagen aqueous solution is liquid state at 4℃,while it is solid state at 37 ℃ .The character coincide with that of collagen.
