Gelatin is a kind of structural protein, widely spreading in bones, tendons andother connective tissue, accounting for70%of the dry matter in some fish skin. Inorder to make the most use of the waste materials from the aquatic products process,the getatin from walleye pollock skin, tilapia skin and donkey skin was extracted.Using the donkey skin gelatin as positive control, the effect of improving Irondeficiency anemia (IDA) of gelatin from walleye pollock skin and tilapia skin isrespectively studied and its mechanism is preliminarily discussed. The main researchresults are shown as follows:It is found that the gelatin from walleye pollock skin and tilapia skin has similaramino acid composition but both slightly different from donkey skin gelatin via theamino acid analysis. The results of mineral analysis showed that the content of Cu,Fe, Zn of donkey skin notablely higher than the other two, which explained the reasonof donkey skin has better blood tonic effect from one side. The tilapia has letter Caand Mg than the other two but its other minerals are closer to the content in donkeyskin gelatin than walleye pollock skin gelatin. The SDS-PAGE(sodium dodecylsulfate polyacrylamide gel electrophoresis)shows that the three kinds of gelatin allconform to the characteristics of typeⅠcollagen. They all consists of three α-strand.The subunit compose of tilapia skin gelatin similar to the gelatin from donkey skin.The gelatin from walleye pollock skin and tilapia skin both has higher gel strengthand lower viscidity than the gelatin from donkey skin, so the two kind of fish gelatinboth can be developed into good bulk gelatin product.The IDA model rats were established by feeding with iron-deprived food and thebleeding via tail vein. Using the donkey skin as positive control, the effect ofimproving IDA of gelatin from walleye pollock skin, tilapia skin and its collaborativeblood tonic effect is studied. The results show: the two kinds of fish skin gelatin bothcan promote the IDA rats’ weight recovery, the increase of the rats’ hemoglobin (Hb) content, the red blood cell count(RBC), the main red blood cell volume(MCV) andthe platelet count(PLT). The gelatin from tilapia skin has better effect than the gelatinfrom walleye pollock skin and the high dose group(500mg/kg) is better than the lowdose group(100mg/kg), the effect is even better if adding FeSO4. The serum iron(SI)content of the rats feeding with high dose of two kinds of fish skin gelatin and FeSO4are significantly improved. It is found from the bone marrow observation that thegelatin from tilapia skin and the walleye pollock skin can recede the phenomenoncaused by IDA and impede the hyperplasia of polychromatophilic normoblast andorthochromatic normoblast.The mRNA expressions of EPOR(erythropoietin receptor), GM-CSF(granulocy-remacrophage colony-stimulating factor) and TPO(thrombopoietin) were investigatedby RT-PCR(reverse transcription-polymerase chain reaction). The results show thatthe gelatin from the two kinds of fish skin can significantly increasing the three kindsof mRNA expression, infering that the gelatin can promote the synthetize andsecretion of EPO, GM-CSF and TPO, stimulate the proliferation and differentiation oferythroid progenitor cell, single progenitor cell and colony-forming unitmegakaryocyte to promote hematopoietic function.The effect of improving IDA of gelatin from walleye pollock skin and tilapiaskin is first investigated and its mechanism is preliminarily discussed, which providescertain scientific proof for depth development and high-value utilization of walleyepollock skin and tilapia skin, while applying to nourishment and health care productsin the future.
