7Compounds were isolated from the Compounds isolated from the extracted substance from seed hulls of Glycinemax(L.)merr by column chromatography of silica gel,ODS,SephadexLH-20and PHPLC ect., whose structures were identified on the basis of spectroscopic analysis including NMR and MS.The compounds were:(1)(+)-catechin,(2)(2-O-(3,4-dihydroxybenzoyl)-2,4,6-trihydroxyphenylacetic acid),(3)(3,5,7,3’,4’-pentahydroxy-6-methoxy-flavone-3-O-β-D-glucoside),(4) quercitin-3-O-β-D-glucoside,(5) phloretin2’-O-a-L-glucopyranoside,(6)2-(2,4-dihydroxy-6-(3,4,5-trihydroxycyclohexanecarbonyloxy)cyclohexyl)acetic acid,(7) Procyanidin B2.A spectrophotometric method to measure the concentration of anthocyanidin from the seed coat of black soybean was established, which use delphinidin as the standard, anthocyanidin was measured with1%HCl acid methanol solution developing agent and detected at520nm wavelength. The linear curve was A=0.0943x (r=0.9996) with the linear in the range of1.7μg/mL-8.5μg/mL for anthocyanidin.The results showed that the contents of anthocyanidin in the seed coat of black soybean from Anhui and Neimenggu were higher than from others.The method in this study was simple, rapid, reliable and accurate. In conclusion, this research has provided rational and experimental basis for further development and quality of this drug.
