Phloretin,3-(4-Hydroxyphenyl)-1-(2,4,6-trihydroxyphenyl)-1-propanone is a flavonoid with broad biological activity, which is present in apples, pears, other fruits and juices in a variety of vegetables. It is named for which is more concentrated in the roots or root bark. Phloretin has many biological and pharmacological activities including antioxidant, whitening, anti-inflammatory, anti-cancer, immune suppression, prevention and treatment of cardiovascular disease and other beneficial organisms to human health. Due to its wide function in health care, drug and high economic value, the further development and use of this resource is important. Currently, only to rely on plant extracts of phloretin has been unable to meet market demand. Therefore, the synthesis and purification of phloretin is significanceIn this thesis, phloretin has been synthesized with new method, the response surface experiments were taken to optimize the conditions and feed ratio in the reaction. We also studied the solid-liquid equilibrium of the reactant and product.The main contents of this thesis are as following:In the first chapter of this thesis, we summarize the references about our research, physicochemical properties, pharmacological effects, biological sources and synthetic methods of phloretin. The research contents of this thesis were described on this basis.In the second chapter, phloretin was synthesized from naringin through a simple two-step reaction. Moreover, the response surface experiments were taken to optimize the conditions and feed ratio in the reaction. The intermediate product and end product were obtained and identified by MS and NMR.In the third chapter, the solid-liquid equilibrium of phloretin was studied by analytical method. Simple procedure to analyze phloretin with high performance liquid chromatography (HPLC) method was set up. The solubilities of phloretin in twelve commonly used solvents and ethanol-water mixed solvents were determined using HPLC at different temperatures.In the forth chapter, the two-parameter, the three-parameter equation and UNIQUAC equation were used to correlate the experimental data of the solubilities of phloretin in different solvent systems and showed satisfactory results.In the last chapter, all of the experimental work was summarized. We have discussed the deficiencies in this thesis and proposed ideas for the latter research.
