In this paper, I have further research about Junzao polysaccharide on the basis of previous studies. Initially,crude polysaccharide from ZizyphusJujuba cv.Junzao (CJP) was purified, subsequently the structural characterization of its purified fractions was elucidated by chemical methods, and then Antioxidant activities of CJP and its purified fractions was studied using chemiluminescence.Main contents and conclusions are as follows:1. Initially, crude polysaccharide was obtained by extraction, followed by deproteination, decolorization, subsequently crude polysaccharide was fractionated by DEAE-Sepharose CL-6B and Sepharose CL-6B. It was applied to a DEAE-SepharoseCL-6B column, one neutral polysaccharide and four acidic polysaccharides were obtained, namely JuNP1, JuSP2, JuSP3, JuSP4, JuSP5, respectively. Its yield were1.9%,13.8%,10.7%,36.1%,13.2%, respectively. JuSP2and JuSP4was eluted by Sepharose CL-6B, and JuSP2, JuSP4were my object of study.Asymmetrical narrowpeakwas obtained on high-performance gel permeation chromatography, andthe specific rotation values of JuSP2b, JuSP4c were constant. These results demonstrated that they were homogenouspolysaccharide. The molecular weight of JuSP2b, JuSP4c were25575Da,14524Da separately.2. The structural characterization of JuSP2b, JuSP4c were investigated by UV, IR and NMR. JuSP2b contained small amounts of protein,and protein-free of JuSP4c. IR spectrum shows JuSP2b and JuSP4c all had characteristic absorption bands of carbohydrates, and IR spectra in the wave number of JuSP4c at827.46cm-1,896.31cm-1could be associated with α-glycosidic linkage and β-glycosidic linkage. The results were consistent with1H NMR spectrum. JuSP2b was composed of Rha, Ara, Xyl, Man, Glc, Gal with the molar ratio of0.23:2.38:0.51:0.39:0.91:3.29, while JuSP4was consisted of Rha, Ara, Xyl, Man, Gal in a molarratio of2.34:9.56:0.85:2.21:2.71.Based on Periodate oxidation-Smith degradation, JuSP2b and JuSP4c may existed in (1â†'),(1â†'2),(1â†'6),(1â†'2,6),(1â†'4)or(1â†'4,6) bond type. 3. Antioxidant activities ofCJP, JuSP2b and JuSP4c were investigated by chemiluminescence. It involved superoxide anion,hydroxyl radical,hydrogen peroxidescavengingactivity. IC50values were obtained in vitro activity tests. The scavengingactivityofCJP, JuSP2b and JuSP4c followed the order:CJP>JuSP4c> JuSP2b.More effective scavengingactivity of CJP than JuSP4cmay be related to other antioxidant (e.g., pigment). Pigment and polysaccharides played a synergistic role, enhancing antioxidant activity. JuSP4c showed a more pronouncedscavengingactivity owing to its more Uronic acid content...
