This paper studied that the application of fluorescein in the light offlow injection chemiluminescence analysis; Synthesis and Characterizationof fluorescein group-containing vinyl monomer and it copolymer; andfluorescein vinyl monomers and acrylamide copolymer applied to flowinjection chemiluminescence analysis. The main work is as follows:(1)The chemiluminescence characteristic of human chorionicgonadotropin(HCG)-fluorescein complex has been studied. Theexperimental conditions for the fluorecein-HCG-ClO-CTMABchemiluminescent system were optimized. Based on the relation betweenchemiluminescence intensity and human chorionic gonadotropinconcentration, a flow injection chemiluminescence assay method has beenestablished for the quantitative determination of HCG. The linear range forHCG is0.8~200mIU/mL. The detection limit is0.01mIU/mL. The relativestandard deviation (n=11) for the determination of10mIU/mL HCG is0.8%. The method was applied to the determination of HCG in the mixedsamples and pregnant women,s urine samples with satisfactory result.(2)Using the method of preparing fluorescent polymer monomerfluorescence function based on fluorescein chromophores, choice, make itreacted with acryloyl chloride, synthesis of fluorescein monomer Ac-Flu containing double-bond copolymerization with acrylamide, then it isprepared, containing fluorescein chromophore water-soluble fluorescentpolymer poly (Ac-Flu-co-AM). The experimentally obtained fluoresceinvinyl monomers and polymers using magnetic (1H NMR), infraredspectroscopy (FT-IR), differential scanning calorimetry (DSC), UV-visiblespectrophotometer (Uv-Vis), fluorescence spectrum method to characterizethe structure and properties.(3)The chemiluminescence characteristic of bovine serum protein(BSA) labelled with fluoresecein has been studied. The experimentalconditions for the fluoresecein-BSA-ClOˉ-CTMAB(cetyltrimethylammonium bromide) chemiluminescent system wereoptimized. Based on the relation between chemiluminescence intensity andBovine serum protein concentration, a flow injection assay method has beenestablished for the quantitative determination of Bovine serum protein.Under the optimized conditions, The linear range for BSA is0.004-8μg/mL,the detection limit is0.04μg/mL; the regression equation wasICL=45.10+479.80C(μg/mL), with a correlation coefficient of0.9998(n=22).The relative standard deviation(n=8)for the determination of2μg/mL BSA is0.6%. And the determination of protein in Qi Chun Jane meters by thismethod, It gets a satisfactory result, the recovery percent is90.0%~106.5%, with using standard addition method to measure its recovery. Result of this experimental shows that the method can be used todetermine the protein from rice.
